摘 要:目的探讨当归补血汤对大鼠局灶性脑缺血再灌注损伤后的神经保护作用以及对海马区一氧化氮合酶(nitric oxide synthase,NOS)表达的影响。方法采用线栓法制成大鼠大脑中动脉缺血再灌注(MCAO)模型,阻断血流2 h后再灌注。应用免疫组织化学染色法检测大鼠局灶性脑缺血再灌注后海马区NOS免疫反应阳性细胞的表达情况,观察当归补血汤对其结果的影响。结果模型组大鼠海马各亚区及齿状回NOS阳性细胞分布稀疏,数目较假手术组和当归补血汤组显著减少,差异有统计意义(P<0.05)。结论大鼠脑缺血再灌注后海马区NOS免疫反应阳性细胞的表达降低,当归补血汤能增强脑缺血再灌注后海马区NOS阳性细胞的表达,对大鼠脑缺血再灌注后的神经组织具有一定的保护作用。Objective To investigate the neuroprotective effect of Danggui Buxue Tang and the effect on the expression of nitric oxide syntbase ( NOS ) in hippocampal region of rats of cerebral isehemia and reperfusion. Methods The rat models of middle cerebral artery ischemia-reperfusion (MCAO) were made by adopting MCAO methed. The blood flow had been blocked for 2 hours and reperfusion. The expression of immunoreactive positive cells of NOS in hippocampal region of rats of cerebral ischemia and reperfusion was detected by immunohistoehemi cal staining method, and observed the effects of Danggui Buxue Tang on the expression of NOS. Results Com pared with sham-operation group and Danggui Buxue Tang group, the positive cells distribution of NOS in hippocam pal region and dentate gyrus of rats of model group were sparse, the number of positive cells was significantly re duced( P〈0.05 ). Conclusion The expression of immunoreactive positive cells of NOS in hippoeampal region of rats of cerebral isehemia and reperfusion was reduced. Danggui Buxue Tang could induce the expression of positive cells of NOS, and has certain protective effect on nerve tissue in rats of cerebral ischemia and reperfusion.
