摘  要:研究了在MES液体培养基内添加0.1~10.0 mg·L-1IAA诱导细基江蓠繁枝变种藻体切段再生芽形成无性系的效果。结果显示,藻体切段只能在形态学上端产生再生芽是细基江蓠繁枝变种的特性。与对照组相比,培养液添加0.1~5.0 mg·L-1低浓度的IAA均能不同程度提高藻体切段切口处再生芽发芽率和生长率,其中添加2.0 mg·L-1IAA对藻段再生芽生长的促进效果最佳,培养至35 d时,再生芽平均长度达到(25.99±1.28)mm,明显高于对照组平均长度(18.50±0.92)mm。然而,培养液添加10.0 mg·L-1高浓度的IAA,反而抑制了藻段再生芽的形成与生长,相比于对照组,再生芽数量最少,发芽率和生长率最低,培养至35 d时,再生芽平均长度仅为(12.23±0.85)mm,低于对照组。研究结果表明,IAA诱导江蓠再生芽的最适宜浓度为2.0 mg·L-1,可为进一步构建细基江蓠繁枝变种种苗快繁应用技术提供基础。Effects of MES liquid medium with the concentrations of O. 1 - 10. 0 mg· L-1 IAA on induction ofregenerated buds from algal fragments in Gracilaria tenuistipitata var. liui Zhang et Xia were studied. The re- suits showed that the morphological upper ends of algal fragments characteristically germinated newborn buds that eventually developed into asexual lines. Compared to the control group, lower concentrations of 0. 1 -5. 0 mg· L-1 IAA added in liquid culture medium all improved the sprouting rates and growth rates of regenerated buds in various degrees, and the promotion effect of 2.0 mg· L-1 IAA was the best, and the mean length of regenerated buds was (25.99 ±1.28 ) mm, obviously higher than that of the control with ( 18.50± 0. 92 ) mm after 35 days of culture. However, the group with concentration more than 10. 0 mg· L-1 IAA restrained form- ing and growth of the regenerated buds, and made them with the lowest quantities, sprouting rates and growth rates, and the mean length of regenerated buds was only( 12.23±0. 85)mm, lower than that of the control group after being cultured for 35 days. It was indicated that the optimum concentration of IAA for inducing re- generated buds of the species was 2.0 mg· L-1. These base conclusions would be benefit for establishing fur- ther applied technique of rapid propagation in G. tenuistipitata var. liu.
