抗U_1RNPSm抗体诱导人淋巴细胞凋亡的研究

摘　　要：研究抗U1 RNP Sm抗体与人淋巴细胞凋亡的关系。方法 :将抗U1 RNP Sm自身抗体与人淋巴细胞共育 ,37℃ 48h后用电镜观察抗体作用细胞凋亡的形态学改变 ;瑞氏 姬姆萨染色观察细胞凋亡率 ,并与流式细胞术进行比较。结果 :电镜观察发现与抗U1 RNP或抗Sm抗体共育后的淋巴细胞发生典型凋亡形态学改变 ,瑞氏 姬姆萨染色计数凋亡率分别为(9.95± 2 .15 ) %和 (15 .72± 3.2 0 ) % ,显著高于对照组 (P <0 .0 1) ,流式细胞术检测结果与形态学方法相符。DNA电泳出现典型梯状条带。结论 :抗U1 RNP Sm抗体具有诱导淋巴细胞凋亡的能力。Objective:Study of the effect of anti-U 1RNP /Sm antibodies on apoptosis of human lymphocytes. Methods: The ultrastructural features of apoptosis were observed with electron microscope and the percentages of apoptosis were measured by Wright-Geamsa staining.The correlation between the results of staining and those of FACS analysis was compared.Results: Ultrastructural observations with electron microscope showed characteristic features of apoptosis and the percentages of apoptosis measured by Wright-Geamsa staining were notably increased compared with the controls〔x±s, (9.95±2.15)% and (15.72±3.20)% versus (2.82±1.19)%, P<0.01〕. There was an excellent correlation between Wright-Geamsa staining approach and FACS analysis.A typical “DNA ladder” was observed by DNA electrophoresis. Conclusion:Anti-U 1RNP/Sm antibodies can induce apoptosis in human lymphocytes.
