摘  要:目的 探讨造血干细胞移植中进行供 受体HLA DRB1高分辨基因分型的重要作用 ,并对HLA DRB1等位基因频率在中国人群中的分布进行分析。方法 采用美国莱姆达公司提供的微量SSPTMHLA DRB1PCR SSP低分辨及高分辨分型试剂盒对 10 1例造血干细胞移植供 受体进行高分辨基因分型 ,其中供 受体 37对 ,无关个体 5 0例。结果 在 37对供 受体分型中 ,有 2对为低分辨HLA DRB1位点相合 ,而高分辨结果为不合 ,其余 35例低分辨及高分辨结果一致。对 5 0例无血源关系供体的HLA DRB1位点高分辨结果进行了等位基因频率分析 ,发现DRB1 0 90 12、DRB1 12 0 2 1/0 2 2、DRB1 0 70 1、DRB1 0 30 11、DRB1 15 0 11/0 12出现频率比较高 ,其频率分别为 36 %、2 2 %、2 0 %、12 %、12 %。结论 HLA DRB1高分辨基因分型分辨率高 ,可明确判定基因型别 ,对于同胞间或无关供Objectives To study the significance of HLADRB1 high resolution typing method in hematological stem cell transplantation and to analyze the distribution of HLADRB1 alleles in Chinese population Methods Low resolution and high resolution typing methods of HLADRB1 PCRSSP (Lambda, Inc) were employed to type 101 cases of related hematological stem cell transplant donors and recipients, in which there were 37 donorrecipient pairs and 50 unrelated individuals Results Among the typing results of 37 donorrecipient pairs, there were 2 pairs whose low resolution typing results were identical, but the high resolution results were incompatible, and the typing results by the two methods in the other 35 pairs were identical HLADRB1 allelic frequencies of the 50 unrelated individuals were analyzed by using HLA high resolution typing results, in which the frequencies of DRB1*09012, 12021/022, 0701, 03011 and 15011/012 were relateivly high and their frequencies were 36%, 22%, 20%, 12% and 12% respectively Conclusions HLADRB1 high resolution typing method can accurately designate HLADRB1 alleles and possesses great importance in related and unrelated hematological stem cell transplantation
