摘  要:目的观察肝X受体(liver-X-receptors,LXRs)对转化生长因子β1(TGF-β1)生物功能的影响,并探讨其可能的机制。方法将培养的人气道上皮细胞分为对照组、TGF-β1组、LXRs低剂量组、LXRs中剂量组和LXRs高剂量组,LXRs组分别予2.5 mol/L、5 mol/L和10 mol/L的LXRs共孵育2 h后,与TGF-β1组一起加TGF-β1反应。2 h后用Real-time PCR方法检测各组细胞纤溶酶原激活物抑制剂1(PAI-1)的mRNA表达,在反应30 min时用免疫荧光法检测P-Smad2的核转位情况。结果 TGF-β1组PAI-1 mRNA表达较对照组明显升高,差异有统计学意义(P<0.05),LXRs组PAI-1 mRNA表达较TGF-β1组明显降低,差异有统计学意义(P<0.05),LXRs剂量越大,降低越明显,予LXRs 10 mol/L干预PAI-1的mRNA下降最明显。对照组、TGF-β1组、LXRs高剂量组P-Smad2阳性细胞百分比分别为(1.752±0.423)%、(95.060±1.854)%、(1.941±0.409)%,对照组与TGF-β1组比较差异有统计学意义(P<0.05),TGF-β1组与LXRs高剂量组比较差异有统计学意义(P<0.05)。结论 LXRs可能通过减少核内P-Smad2含量来影响TGF-β1的生物学功能。Objective To observe effect of LXRs on TGF-β1 in bronchial epithelial cells in order to find the possible mechanisms.Methods Cultural human bronchial epithelial(HBE) cells were divided into control group,TGF-β1 group,low-dose of LXRs group,moderate-dose of LXRs group and high-dose LXRs group.LXRs groups were incubated with different doses of LXRs(2.5 mol/L,5 mol/L and 10 mol/L) for 2 hours.Then LXRs groups and TGF-β1 group were incubated with TGF-β1.After 2 hours,mRNA expressions of cellular plasminogen activator inhibitor 1(PAI-1) in each group were detected by Real time PCR method.After being incubated with TGF-1 for 30 minutes,P-Smad2 nuclear translocation was detected by immumofluorescence method.Results The expression of PAI-1 mRNA in TGF-β1 group was significantly increased compared with that of control group(P0.05);the expression of PAI-1 mRNA in LXRs groups were significantly lower than that of TGF-β1 group(P0.05);the larger LXRs dose was,the more obvious reduced and the most mRNA descent of PAI-1 were in LXRs 10 mol/L group.P-Smad2 positive cell percentage in control group,TGF-β1 group and high-dose LXRs group were(1.752±0.423)%,(95.060±1.854)%,and(1.941±0.409)% respectively,and the difference was statistically significant between control group and TGF-β1 group(P0.05).The difference was also statistically significant between TGF-β1 group and high-dose LXRs group(P0.05).Conclusion LXRs may affect biological function of TGF-β1 by attenuated P-Smad2 phosphorylation.
