摘  要:自然环境中生长的桑树经常会遭遇干旱等不良环境胁迫,从蛋白质水平探究桑树的抗旱性形成机制,对于指导桑树抗旱品种选育具有重要意义。以桑树品种新一之的幼苗为材料,经含5%聚乙二醇(PEG)6000的Hoagland营养液模拟干旱胁迫处理后,应用蛋白质双向凝胶电泳(2-DE)技术检测桑树幼苗叶片的蛋白质表达差异,共有45个蛋白点发生了显著变化:与正常环境对照组相比,干旱胁迫处理组中有25个蛋白点上调表达,包括8个特异性表达的蛋白点,有20个蛋白点下调表达,其中的12个蛋白点仅存在于对照组。利用MALDI-TOF-TOF-MS技术鉴定差异明显的蛋白点,发现其中3个蛋白分别与过氧化物清除、甲硫氨酸氧化还原反应和脂多糖的合成有关,有1个蛋白包含保守结构域UBN2和DUF1664,这些蛋白点的表达变化可能与干旱胁迫密切相关。Mulberry(Morus L.) growing in natural environment frequently confronts unfavorable environmental stress such as drought.To explore the formation mechanism of drought resistance in mulberry at protein level is of important significance to guide the breeding of drought resistant mulberry variety.In this study,the mulberry variety Neo-Ichinose was used as material to detect differentially expressed proteins in seedling leaves by two-dimensional electrophoresis(2-DE) technology after drought-stress treatment with Hoagland nutrient solution containing 5% polyethylene glycol(PEG) 6000.The results showed that 45 protein spots displayed significant alterations.Compared with the control group under normal environment condition,the drought-stress treated group had 25 up-regulated(including 8 specifically expressed) protein spots and 20 down-regulated protein spots,among which 12 protein spots were only found in the control group.Differential proteins which had significant differences were identified with MALDI-TOF-TOF-MS.It was found that 3 protein spots are related to peroxide elimination,methionine redox reaction and lipopolysaccharide biosynthesis respectively,among which 1 protein spot contains the conserved domains UBN2 and DUF1664.The expressional variation of these protein spots may be closely related with drought stress.
