摘　　要：采用垂直板状聚丙烯酰胺梯度凝胶电泳技术,分析了乌龟(Chinemys reevesii)的心、脑、眼、肝、胃、肺、小肠、肌肉、脾、性腺10种组织中的酯酶(EST)、乳酸脱氢酶(LDH)、苹果酸脱氢酶(MDH)和乙醇脱氢酶(ADH)4种同工酶的表达模式,并对其同工酶位点及酶谱表型进行了分析。结果表明,乌龟4种同工酶系统具有不同程度的组织特异性。在心、肝脏、肺、小肠、肌肉和性腺6种组织中检测到EST活性,并在肝脏中检测到7条酶带;LDH酶带多于典型的5条,其中Ldh-1在胃和脾中被检测到,而Ldh-2只在脾中被检测到;MDH具有上清液型(s-MDH)和线粒体型(m-MDH)两种类型,s-MDH型的活性高于m-MDH的活性;ADH中检测到3条酶带,由两个基因位点编码。同工酶的种类与活性的变化与其功能相一致。Four isozymes（EST,LDH,MDH and ADH）from ten tissues（heart,brain,eye,liver,stomach,lung,small intestines,muscle,spleen and gonad） of Chinemys reevesii from Poyang Lake were analyzed by vertical polyacrylamide gradient gel electrophoresis.The results revealed that the four isozymes had different degrees of tissues specificity.EST was only expressed in six tissues including heart,liver,lung,small intestines,muscle and gonad.Seven bands of EST were identified in liver.LDH enzymatic had more than five typical bands.Ldh-band was identified in spleen and stomach,but Ldh-2 band was just identified in spleen.The types of MDH including m-MDH and s-MDH could be identified in all tissues.The activities of s-MDH was found to be higher than m-MDH.Three bands in ADH were identified and encoded by two loci.The changes of their activities were consistent with physiological functions.
