摘 要:从四川省两个不同肉鸽养殖场的送检病鸽中分别分离出2株鸽源新城疫病毒(NDV),分别命名为Mianyang12505和sms12。致病性试验表明2株鸽源NDV均为弱毒,但融合蛋白(F)基因序列分析表明2个毒株均含有典型的强毒株F蛋白裂解位点112RRQKRF117;以F基因序列为基础构建遗传发育树,发现2株鸽源NDV属于基因Ⅵ型;F基因突变分析表明,鸽源NDVF基因突变率较大,且基因Ⅵ型和Ⅶ型存在规律突变;核酸和氨基酸序列同源性分析表明,国内鸽源NDV毒株核酸序列同源性为83.5%~99.7%,氨基酸同源性为85.6%~100%,且本研究分离的毒株与JS0722Pi同源性最高。Two Newcastle disease virus (NDV)strains were isolated from sick pigeons in Sichuan province,China,and were designated as Miangyang12505 and sms12, respectively. The result of pathogenicity test showed that two NDVs were lentogenic isolates, while analysis of Fusion protein gene sequence demonstrated that these two pigeon-origin NDVs contained a classical motif of 112RRQKRF117 that often appeared at the cleavage site of F gene of velogenic NDVs. The phylogenetic tree, which showed that Mianyang12505 and sms12 belonged to genotype Ⅵ ,was constructed based on the sequence of F gene. Analysis of mutations of F gene indicated that the mutation rate was very high among different pigeon-origin NDVs and there was a regular substitution in genotype Ⅵ and genotype Ⅶ. Homology analysis of nucleotide and deduced amino acid of F gene showed that nucleotide homology of F gene ranged from 83.5% to 99.7% and amino acid homology of F gene from 85.6% to 100%,and the F genes of two viruses in this study were the most homologous to the JSO722Pi's.
