高效液相色谱法测定复方消渴滴丸中葛根素含量

摘　　要：目的建立复方消渴滴丸中葛根素的含量测定方法。方法采用高效液相色谱法,色谱柱为C18柱(200 mm×4.6 mm,5μm),流动相为甲醇-水(28∶72),流速1.0 mL/min,柱温30℃,紫外检测波长250 nm,进样体积10μL。结果葛根素质量浓度在83.68～836.8μg/mL内与峰面积线性关系良好,r=0.999 8(n=7)。高、中、低3种不同质量浓度溶液的平均回收率为99.08%,RSD=1.09%(n=9)。结论该方法简便易行、重复性好,可作为复方消渴滴丸的质量控制方法。Objective To establish an HPLC method for the determination of assay and of puerarin in Fufangxiaoke Pills. Methods The chromatographic conditions were C18 columne quipped with a C18 column（200 mm ×4.6 mm,5 μm）, with the mobile phase of methanol ： water（28：72）,the flow rate was 1.0 mL/min and the detective wavelength was 250 nm, the injection volumn was 10μL. Results The puerarin curves were linear over the concentration range of 83.68-836.8 μg/mL（RSD=0.999 8）,and the average recovery was 99. 08% for three different levels of the added amount of puerarin, RSD = 1.09% （n = 9）. Conclusion The method is simple and reproducible. It can be used as the preparation of quality control methods.
