摘 要:目的探讨醒脑开窍针刺法干预缺血再灌注后脑组织炎症反应的分子机制。方法以醒脑开窍针刺法为干预手段,以大鼠急性局灶性脑缺血再灌注模型作为研究对象,运用RT-PCR方法,观测不同时间点病变侧大脑皮层2种关键炎性细胞因子受体即白细胞介素-1受体Ⅰ(IL-1RI)和肿瘤坏死因子受体Ⅰ(TNFR-Ⅰ)的mRNA表达情况,并观察针刺对上述指标的调节作用规律。结果醒脑开窍针刺法降低了大鼠脑缺血再灌注后大脑皮层异常升高的IL-1RI、TNFR-Ⅰ的mRNA的表达,且该作用随时间的延长而更为明显。结论醒脑开窍针刺法可干预缺血再灌注后脑组织炎症反应,进而对脑损伤起保护作用。OBJECTIVE To research the cellular and molecular mechanisms of resuscitating acupuncture against inflammatory reaction in brain after cerebral ischemia reperfusion.METHODS Acupuncture method of XNKQ(activating brain and opening orifices) were applied as therapeutic method,and the rat model of acute focal cerebral ischemia reperfusion as research object.The technology of RT-PCR was used to observe the mRNA expression of two key inflammatory cytokine receptors(IL-1RI and TNFR-Ⅰ) in different times after cerebral ischemia reperfusion.The regulatory law of acupuncture on the above indexes was also observed.RESULTS XNKQ acupuncture reduced the mRNA expression of IL-1RI and TNFR-Ⅰ in brain tissues of rats with cerebral ischemia reperfusion.The role of acupuncture was more obvious as the time past.CONCLUSION The acupuncture method of XNKQ could interfere inflammatory response in brain after cerebral ischemia reperfusion,and thus plays a protective role against brain injury.
