大黄素对人胃癌MNK-45和MGC8-03细胞增殖的影响

摘　　要：目的观察大黄素对人胃癌MNK-45和MGC8-03细胞增殖的影响。方法采用MTT法测定MNK-45细胞和MGC8-03细胞增殖抑制率;采用AO/EB双染观察细胞形态学变化;采用流式细胞仪测定细胞周期。结果大黄素(5～80μg/mL)对MNK-45和MGC8-03细胞增殖均具有显著抑制作用(P<0.01或0.05),且药物剂量越大,作用时间越长,其抑制作用越强(P<0.01);大黄素作用于两种细胞后荧光显微镜下观察均显示细胞凋亡特征;大黄素使MNK-45和MGC8-03细胞增殖被阻滞于G0/G1期。结论大黄素能抑制人胃癌MNK-45和MGC8-03细胞增殖,诱导其凋亡,影响其细胞周期时相的分布。Objective To observe the effects of emodin on the proliferation of human gastric carcinoma cell lines MNK-45 and MGC8-03.Methods MTT assay was used to measure the inhibitory rate on the cell growth of MNK-45 and MGC8-03.The morphological changes were determined using AO/EB staining.The changes of cell cycle were detected by flow cytometry.Results Emodin（5―80μg/mL） had the significant inhibition on cell proliferation of MNK-45 and MGC8-03（P0.01 or 0.05）.In addition,the higher the dosage was,the longer the effective time was,and the stronger the inhibiton was（P 0.01）;After emodin treatment,the two cell lines showed the features of apoptosis;Emodin could make the cell proliferation of MNK-45 and MGC8-03 stay in the G0/G1 phase.Conclusion Emodin could inhibit the cell proliferation of human gastric carcinoma MNK-45 and MGC-803,induce the apoptosis of MNK-45 and MGC-803,and affect the distribution of cell cycle phase.
