补肾填精法对再生障碍性贫血模型大鼠调控因子影响的实验研究

摘　　要：目的观察补肾填精法对再生障碍性贫血(aplastic anemia,AA,简称再障)模型大鼠调控因子的影响。方法将清洁级Wistar大鼠36只随机分为对照组、模型组、治疗组(补肾填精中药)3组各12只;对照组正常饲喂,模型组和治疗组用5-氟尿嘧啶(5-FU)与马利兰合用诱导建立大鼠AA模型,之后治疗组每天给予补肾填精中药10g/kg-1·d灌胃,连续4周,4周后观察各组大鼠外周血WBC、RBC、Hb、PLT及调控因子的变化。结果①模型组与治疗组外周血以上各值水平显著低于对照组,治疗后治疗组外周血以上各值水平提高,差异有统计学意义;②模型组与治疗组调控因子白介素-2(Interleukin-2,IL-2)、肿瘤坏死因子-α(Tumor necrosis factorα,TNF-α)含量显著高于对照组,治疗后治疗组IL-2、TNF-α含量下降,差异有统计学意义。结论补肾填精法能明显改善再障模型大鼠的骨髓造血功能,并可有效改善细胞因子分泌异常,促进骨髓造血功能的恢复。Objective To observed the effect of Aplastic Anemia the model rat's Regulation Factors Treated with Tonifying Kidney and Replenishing Essence Method.Methods 36 Clean-class Wistar rats,were randomly divided into 3 groups,namely control group,model group,treatment group(TKRE prescription).The control group was fed normal foods,model group and treatment group were given the 5-FU plus Myleran to induce AA model of rat.After that,the treatment group rat were fed with TKRE prescription 10g /(kg·d) for 4 weeks,later,to observe the numbers of blood cells and IL-2 TNF-α of each guoup.Results ① the numbers of blood cells in model group and treatment groups were significantly decreased than the control group,the numbers of blood cells in treatment group was higher after treated,the difference was statistically significant.② IL-2 and TNF-α levels were significantly elevated in model group and treatment groups than the control group,after treated,IL-2 and TNF-α levels were declined in treatment group,the difference was statistically significant.Conclusions From point view of above,the tonifying kidney and replenishing essence method could significantly improve bone marrow hematopoietic function of aplastic anemia model of rat and effectively improve abnormal secreteing of regulation factors and encouraged the bone marrow hematopoietic function's recover.
