骨髓间充质干细胞肝包膜下移植治疗大鼠急性肝损伤

摘　　要：目的了解大鼠骨髓干细胞对大鼠急性肝功能损伤的治疗作用。方法用四氯化碳腹腔注射法建立大鼠急性肝损伤模型,将大鼠骨髓间充质干细胞注射移植于模型鼠肝包膜下,比较移植前和移植3周后,血清Alb、ALT、AST水平变化。以0.9%氯化钠溶液肝包膜下注射作对照组。结果干细胞移植3周后,移植组血清Alb水平为(19.2±2.6)g/L,明显高于对照组相应水平(12.6±3.5)g/L,(P<0.05);移植组血清ALT和AST水平分别为(20.9±5.1)和(19.4±3.7)U/L,明显低于对照组相应水平(40.7±11.6)和(37.9±12.7)U/L,(P<0.05)。结论大鼠骨髓干细胞肝包膜下移植可使急性肝损伤大鼠的肝功能得到改善。Objective To study the therapeutic effect of rat bone marrow mesenchymal stem cells transplanted under capsule of liver in rat with liver injury. Methods The model rat with acute liver injury was established by intra- abdominal injection of carbon tetrachloride（CCL）. The rat bone marrow stem cells were transplanted into the lacuna of subcapsule（Glisson＇s capsule）of liver in rat. The therapeutic effects were evaluated by＇comparing the levels of albumin （Alb） and aminotransferase（ALT, AST） between the stem cell transplant group and the control group in 3 weeks of transplantation. Results Level of Alb of the transplant group （19.2 ± 2.6） g/L was higher than that of control （12.6 ± 3.5 ）g/L significantly （P〈0.05）. Levels of ALT and AST of the transplant group （20.9 ± 5. 1）, （19.4 ± 3.7） IU/L were significantly lower than that of control （40. 7 ± 11.6）, （37.9 ± 12. 7） IU/L （P〈0.05）. Conclusion The bone marrow mesenchymal stem cells transplanted in subcapsule of liver could improve the liver function in rat with acute liver injury.
