摘  要:目的探讨肌肽(carnosine)对高糖诱导的大鼠心肌成纤维细胞(cardiac fibroblasts,CFs)增殖的影响及作用机制。方法以CFs为研究对象,采用WST-1法观察肌肽抑制高糖诱导CFs的增殖;乳酸脱氢酶(LDH)法检测肌肽对细胞膜的毒性作用;Western blot法观察细胞周期蛋白P21及MAPK通路相关蛋白P38、P-p38的表达水平。结果肌肽显著抑制了高糖诱导的CFs增殖,在5~30 mmol/L范围内呈一定的浓度依赖效应,且与p38 MAPK信号通路相关。结论肌肽可通过P38MAPK信号通路抑制高糖诱导的CFs增殖。Objective To study the effect of camosine on cell proliferation in rat cardiac fibro blasts (CFs) induced by high glucose and the possible mechanism. Methods CFs prolifera tion induced by high glucose was observed by WST1 assay. The eytotoxicity was detected by lactate dehydrogenase (LDH) assay. In addition, Western blot method was used to observe the expression level of cell cycle protein P21, P38 and Pp38. Results Camosine significantly in hibited proliferation of CFs induced by high glucose in concentrationdependant way within 5 - 30 mmol/L. The inhibition of eamosine was closely related to MAPK pathways. Conclusion Carnosine can inhibit proliferation of CFs by high glucose. The mechanism on eamosine' s inhi bition could be related to MAPK pathways.
