摘 要:目的观察血管紧张素Ⅱ(angiotensinⅡ,AngⅡ)对血管内皮细胞内[Ca2+]i和线粒体膜电位的影响及辛伐他汀的保护作用。方法将血管内皮细胞分为空白对照组(仅给予细胞培养液)、单纯AngⅡ组(细胞培养液中加入AngⅡ,使其终浓度为10-7mol/L)、AngⅡ+小剂量辛伐他汀组(在单纯AngⅡ组的基础上加入辛伐他汀,使辛伐他汀的终浓度为0.2μmol/L)、AngⅡ+大剂量辛伐他汀组(在单纯AngⅡ组的基础上加入辛伐他汀,使辛伐他汀的终浓度为2μmol/L),用激光共聚焦显微镜测量各组细胞的[Ca2+]i和线粒体膜电位水平。结果①AngⅡ组的线粒体膜电位显著低于空白对照组(P<0.01);AngⅡ+辛伐他汀组的线粒体膜电位显著高于AngⅡ组(P<0.01);AngⅡ+大剂量辛伐他汀组的线粒体膜电位水平高于AngⅡ+小剂量辛伐他汀组(P<0.05)。②AngⅡ组的[Ca2+]i显著高于空白对照组(P<0.01);AngⅡ+辛伐他汀组的[Ca2+]i显著低于AngⅡ组(P<0.01);AngⅡ+大剂量辛伐他汀组的[Ca2+]i低于AngⅡ+小剂量辛伐他汀组(P<0.05)。结论AngⅡ可引起血管内皮细胞内[Ca2+]i的显著增高及线粒体膜电位的显著降低,而辛伐他汀可逆转AngⅡ的这一作用。Objective To investigate the effects of angiotensin 11 (Ang 11 ) on [ Ca2+ ]i and mitochondria membrane potential in vascular endothelial cell (VEC) and protection of simvastatin. Methods Cultured VECs were divided into 4 groups, control group (only given cell culture medium), Ang Ⅱ group ( Ang Ⅱ was given with the last concentration be 10-7 mol/L), Ang Ⅱ+ low dosage simvastatin group ( simvastatin was given with the last concentration be 0. 2 μmol/L at the base of Ang Ⅱ group, and Ang Ⅱ +high dosage simvastatin group ( simvastatin was given with the last concentration at 2 μmol/L on the base of Ang Ⅱ group). Intracellular free calcium concentration and mitochondria membrane potential were measured by laser scanning confocal microscopy. Results ①Mi- tochondria membrane potential of Ang Ⅱ group was significantly lower than that of control group( P〈 0. 01 ). Mitochondria membrane potential of Ang Ⅱ +simvastatin group was significantly higher than that of Ang II group( P〈0. 01 ). Mitochondria membrane potential of Ang Ⅱ +simvastatin group was higher than that of Ang Ⅱ group ( P〈0. 05 ). ②Intracellular free calcium concentration of Ang Ⅱ group was significantly higher than that of control group(P〈0. 01 ). Intracellular free calcium concentration Ang Ⅱ+ simvastatin group was significantly lower than that of AngⅡ group( P〈0. 01 ). Intracellular free calcium concentration in Ang Ⅱ +high dosage simvastatin group was lower than that of Ang Ⅱ+low dosage group (P〈0.05). Conclusion Ang Ⅱ induce obvious changes of intracellular free calcium concentration and mitochondria membrane potential in VEC, and simvastatin treatment can inhibit it.
