高效液相色谱法测定乳房炎涂膜剂中咖啡酸的含量

摘　　要：为了建立乳房炎涂膜剂中咖啡酸的反相高效液相色谱分析方法,试验采用Waterssymmetry slieldTMC18色谱柱(250 mm×4.6 mm,5μm),甲醇-1%冰醋酸水溶液(20∶80)流动相等度洗脱,流速为1.0 mL/min,检测波长323 nm,柱温为35℃进行检测。结果表明,咖啡酸在8～40μg/mL范围内线性关系良好(r2=0.9989)。平均回收率为100.1%,RSD为0.8%。该方法简便,准确,可用于测定乳房炎涂膜剂中咖啡酸的含量。A method was established for the determination of caffeic acid in mastitis plastic by reversed phase high performance liquid chromatography（RP-HPLC）.Chromatography was performed on a Waters symmetry shieldTMC18 column（250 mm×4.6 mm,5 μm） at 323 nm and 35 ℃.The mobile phase consisted of a mixture of methanol-acetic acid（20∶ 80） at a flow rate of 1.0 mL/min.The results showed that there were good linear relations between the concentrations and the peak-areas of caffeic acid with in the range of 8～40 μg/mL.The standard solution was stable in 16 h（RSD=0.6 %）.The average recovery was 100.1 %（RSD=0.8 %）.This method was simple,accurate,and can be used for the determination of caffeic acid in mastitis plastic.
