摘 要:目的探讨急性应激抑郁模型大鼠大脑中缝核、海马、前额叶、纹状体及外周血5.羟色胺1A(5-HT1A)与5-羟色胺2A(5-HT2A)受体mRNA表达的变化。方法将雄性SD大鼠分为实验组和对照组,每组8只;实验组给予连续3d电击诱导急性应激抑郁模型。采用旷场实验评定其行为学改变;应用逆转录聚合酶链反应检测并比较2组大鼠大脑中缝核、海马、前额叶、纹状体及外周血粒细胞5-HT1A与5-HT2A受体mRNA的表达水平。结果(1)实验组5-HT2A受体mRNA表达在海马、中缝核、前额叶、纹状体分别为0.35±0.29、0.22±0.16、0.79±0.33、0.2±0.18,对照组分别为2.00±2.14、1.05±0.88、1.64±1.51、1.26±0.75,2组在海马、中缝核、纹状体的差异有统计学意义(P均〈0.05)。(2)实验组5-HT1A受体mRNA表达在海马、中缝核、前额叶、纹状体分别为2.02±1.91、0.87±1.54、1.74±1.45、1.52.±1.56,对照组分别为0.99±0.63、0.97±1.34、1.05±1.04、2.48±3.82,2组比较差异无统计学意义(P〉0.05)。(3)实验组外周血粒细胞5-HT1A、5-HT2A受体mRNA表达分别为1.55±0.75和0.47±0.47,对照组分别为1.13±0.34和1.31±1.20,2组比较差异无统计学意义(P〉0.05)。结论急性应激大鼠大脑5-HT2A受体mRNA表达在中缝核、海马、纹状体减少,在前额叶、外周咀无变化;5-HT1A受体mRNA表达在上述脑区及外周血中均无变化。Objective To investigate the message ribonucleic acid (mRNA) expression of 5-HTIA receptor and 5-HT2A receptor in raphe nuclei, hippocampi, prefrontal cortex, striatum and blood of acute stressed depression rat model. Methods All 16 male SD rats were divided into 2 groups: control and experimental group. The experimental group was made by foot shock once a day last for 3 days. Open field was used to measure rats' behavior. The mRNA expressions of 5-HTIA and 5-HT2A receptors in raphe nuclei, hippocampus, prefrontal cortex, striatum and blood were assayed with real-time reverse-transcription polymerase chain reaction (PCR). Results ( 1 ) Compared to control, mRNA expression of 5-HT2A receptor reduced in hippocampi(0. 35 ±0. 29 vs. 2. 00 ±2. 14; P 〈0. 05) , raphe nuclei(0. 22 ±0. 16 vs. 1.05 ± 0. 88 ; P 〈 0. 05 ) and striatum(0. 24 ± 0. 18 vs. 1.26 ± 0. 75 ; P 〈 0.01 ). (2) Compared to control, mRNA expression of 5-HTIA receptor in hippocampi(2. 02 ± 1.91 vs. 0. 99 ±0. 63) , raphe nuclei(0. 87 ± 1.54 vs. 0. 97 ± 1.34) ,prefrontal cortex( 1.74 ± 1.45 vs. 1.05 ± 1.04) , striatum( 1.52 ± 1.56 vs. 2. 48 ± 3.82) of acute stressed depression rat model was not significantly different ( All P 〉 0.05 ). ( 3 ) The blood mRNA expression of both 5-HTIA receptor ( 1.55 ± 0. 75 vs. 1.13 ± 0. 34 ) and 5-HT2A receptor (0. 47 ± 0.47 vs. 1.31 ± 1.20 ) were not significantly different between control and experimental group ( All P 〉 0. 05 ). Conclusion The mRNA expression of 5-HT2A receptor may be decreased in hippocampi, raphe nuclei and striatum of acute stressed depression rat model ,while expression of 5-HT1A receptor might not change.
