摘 要:目的探讨三七总甙对糖尿病皮肤溃疡(DSU)大鼠肿瘤坏死因子-α(TNF-α)的影响。方法 DSU模型制作成功的Wistar大鼠60只随机分为对照组(生理盐水组)和三七总甙高剂量组(200 mg.kg-1.d-1)、中剂量组(100 mg.kg-1.d-1)、低剂量组(50 mg.kg-1.d-1),灌胃30 d,右心房取血,放射免疫法检测血清TNF-α水平,反转录聚合酶链式反应、蛋白质印迹法检测溃疡组织中TNF-αmRNA及蛋白表达。结果与对照组比较,三七总甙高、中、低剂量组血清TNF-α水平随三七总甙剂量的升高而降低(P<0.05),低剂量组高于中剂量组(P<0.05),中剂量组高于高剂量组(P<0.05)。与对照组比较,三七总甙高、中、低剂量组溃疡组织TNF-αmNRA及蛋白表达均下调,低剂量组表达高于中、高剂量组(P<0.05),中剂量组高于高剂量组(P<0.05),差异均有统计学意义。结论三七总甙可降低DSU大鼠TNF-α的表达,在一定剂量范围内可有效控制DSU炎症进展。Objective To investigate the influence of panax notoginsenoside(PNS) on tumor necrosis factor-α(TNF-α) in rats with diabetic skin ulceration(DSU).Methods Sixty Wistar rats were used to build DSU model,then were randomly divided into control group,high-dose PNS(200 mg·kg-1·d-1)group(PNS-H group),middle-dose PNS(100 mg·kg-1·d-1) group(PNS-M group) and low-dose PNS(50 mg·kg-1·d-1) group(PNS-L group).The rats were fed with PNS by intragastric administration for 30 days.The level of TNF-α in serum was detected by radioimmunoassay,the expressions of TNF-α mRNA and protein in ulcer tissue were detected by reverse transcription-polymerase chain reaction and Western blotting methods.Results From the PNS-H group to PNS-L group,the level of serum TNF-α decreased gradually,which in PNS-H group decreased more than in PNS-M group(P0.05) and in PNS-M group decreased more than in PNS-L group(P0.05).Compared with control group,the expressions of ulcer tissue TNF-α mRNA and protein in PNS-H,PNS-M and PNS-L group decreased,the expressions in PNS-L group were significantly higher than those in PNS-H and PNS-M group(P0.05),and the expressions in PNS-M group were higher than that in PNS-H group(P0.05).Conclusion PNS can decrease the expression of TNF-α in rats with DSU,and effectively control the progression of inflammation.
