摘 要:目的选用适宜的三维支架在体外构建类似于体内的肝干细胞(HSCs)生长的微环境。方法以壳聚糖、硫酸肝素和透明质酸三种材料为原料,以冷冻干燥法制孔技术制备壳聚糖-硫酸肝素-透明质酸(C-HS-Ha)三维(3D)多孔隙复合支架,检测该支架的吸水率、孔隙率、孔径及降解率后,接种HSCs注入此支架,通过检测细胞在支架上的黏附率、细胞在支架内的形貌以及生长活力等情况来评价支架。结果吸水率、孔隙率、孔径及降解率都适合细胞生长;壳聚糖-透明质酸(C-Ha)支架与C-HS-Ha复合支架相比较,细胞黏附率前者约为后者的50%,细胞增殖数约是后者的75%;HSCs在支架内相互连接并贯穿孔间交织成网状。结论 C-HS-Ha复合支架是一种适于HSCs生长和增殖的材料,有望作为体外三维环境的支架材料并被用于组织工程和药物筛选等。Objective To study a suitable three-dimensional(3D) porous scaffold for constructing similar in vivo environment for hepatic stem cells(HSCs) culture.Methods This study used three types of bio-materials,chitosan,heparin sulfate and hyaluronate to construct a new chitosan-heparin sulfate-hyaluronate(C-HS-Ha) hybrid three dimensional(3D) porous scaffolds by lyophilization.Selected the pore size,porosity,water absorption and degradation rates for measuring the characters of the scaffold,culturing hepatic stem cells(HSCs) on the scaffold to evaluate the adhesion rate of cells,cellular proliferation and cell activity for assessing the status of HSCs in the C-HS-Ha scaffolds.Results The results showed that: pore size and porosity,water absorption,degradation rates were suitable for cells growing and proliferation.Taking chitosan-glutin(C-G) scaffolds as control,the adhesion rate is 50% and the multiple of proliferation is 75% as C-G scaffolds compared with C-HS-Ha scaffold.The cells interact may grow into network throughout the scaffolds holes that keeping the character of HSCs and proliferating into clusters morphology.Conclusion C-HS-Ha scaffold is suitable for HSCs growing,proliferating and maintaining the characteristics of stem cells,and it can be expected to be novel therapeutic materials for constructing three-dimensional scaffold in similar 3D environment in vitro which enable C-HS-Ha scaffold for tissue engineering as well as drug screening.
