摘 要:珍珠矮根状茎在添加不同质量浓度柠檬酸(0、0.1、0.2、0.5、1.0、2.0、5.0 g/L)的分化培养基(MS+NAA 0.5mg/L+蔗糖30.0 g/L+TDZ 0.1 mg/L)中培养50 d后,统计根状茎的平均出芽数,检测与褐变相关的酶活性和总酚含量.结果表明:柠檬酸质量浓度在0.1~1.0 g/L的范围内,随着培养基中柠檬酸含量的提高,根状茎的出芽数逐步升高;PPO、POD、PAL活性及总酚含量逐步降低,SOD和CAT活性逐步提高,当柠檬酸质量浓度为1.0 g/L,平均出芽数为4.48,培养基未明显褐变;柠檬酸质量浓度在1.0~5.0 g/L的范围内,随着柠檬酸质量浓度的提高,出芽数逐步下降,PPO、POD、PAL活性及总酚含量逐步升高,SOD、CAT活性逐步下降.The rhizomes of Cymbidium nanulum Y. S. Wu et S. C. Chen were incubated on differentiation medium MS + NAA 0. 5 mg/L + sucrose 30. 0 g/L + TDZ 0. 1 mg/L containing citric acid (CA) (0,0. 1,0. 2,0. 5,1.0, 2. 0,5.0 g/L) for 50 days. With the increase of CA concentration ranging from 0 to 1.0 g/L, the activities of PPO, POD, PAL and the total phenol content were decreased gradually, the activities of SOD, CAT and the differ- entiation rate were increased gradually. Whereas, within 1.0 g/L to 5.0 g/L, the activities of PPO, POD, PAL and the total phenol content were increased gradually, the activities of SOD, CAT and the differentiation rate were decreased gradually. Compare to control, supplemented with appropriate concentration of CA on medium can re- duce browning degree and promoted rhizome differentiation rate, the most appropriate concentration of CA was 1.0 g/L CA and the differentiation rate reached 4. 48.
