腐植酸对芘的生物毒性效应的影响

摘  要:本文旨在研究水体中腐植酸(humic acid,HA)对芘胁迫下鲫鱼肝脏中富集芘,活性氧的诱导及氧化损伤的影响.通过静态模拟实验,在水体中不同浓度(0、0.05、0.1、0.5、1.0和2.5mg/L)的HA与芘(0.05mg/L)共同暴露10天后,取鲫鱼肝脏,分析其中芘的富集量、活性氧自由基的诱导信号、脂质过氧化产物丙二醛(MDA)含量以及抗氧化系统发生的变化.结果发现,随着HA浓度的增加,芘在鲫鱼肝脏中的富集降低;0.05mg/L和0.1mg/L的HA显著诱导鲫鱼肝脏产生活性氧自由基,但随着HA浓度升高,这种诱导效应降低;0.5mg/L和1.0mg/L HA可以使肝脏中MDA含量显著降低;三种抗氧化系统酶超氧化物歧化酶,过氧化氢酶,谷胱甘肽-S-转移酶活性及谷胱甘肽含量因HA的加入均显示先升高后降低的趋势.本文认为,HA降低芘在鲫鱼肝脏中的富集,并改变芘对鲫鱼的生物效应,减轻芘对鲫鱼产生的氧化损伤.In the current study, whether the different concentrations of humic acid (,HA)may influence bioaccumulation and oxidative stress of pyrene in liver of Carassius auratus was investigated. The fish(C, auratus) was exposed to HA(0,0.05,0.1,0.5,1.0 and 2.5 mg/L)and pyrene(0.05 mg/L)mixtures for 10 days. Then the pyrene bioaccumulation as well as their effects on the reactive oxygen species, Malondialdehyde(MDA)content and antioxidative system in the liver of C. auratus were analyzed. With the HA concentration increase, bioaccumulation of pyrene in liver of C. auratus reduced. In the meantime,0.05 mg/L and 0.1 mg/L HA could induce the production of reactive oxygen species in the liver of C. auratus,O. 5 mg/L and 1.0 mg/L HA could decrease the MDA contents.The superoxide dismutase, catalase, glutathione-s transferase activilies and glutathione contents decreased after an initial increase, under HA concentration increase. It was pyrene stress which caused the oxidative damage of the liver and brain,which could be further induced by HA. Overall, our present work illustrated that the presence of HA could reduce pyrene concentration in fish liver and change the pyrene's biological effects, which should not be disregarded when considering the ecological safety of pyrene in the aquatic system.
