摘 要:目的观察不同日龄新生鼠脑一氧化氮 (NO)含量 ,一氧化氮合酶 (NOS)活性的变化及甲状腺激素对nNOS基因表达的调节。 方法采用丙基硫氧嘧啶 (PTU)灌注孕母鼠造成甲减模型。采用NO、NOS的生化测定法以及半定量逆转录聚合酶链反应 (RT -PCR)。 结果生化测定法显示 ,不同年龄段大鼠随着年龄的增加NO含量上升 (P <0 .0 1) ,NOS的活性上升 (P <0 .0 1) ;同一年龄段仔鼠 ,甲减组较正常组NO含量下降 (P <0 .0 1) ,NOS活性下降 (P <0 .0 1) ;RT -PCR测定显示 ,幼鼠nNOSmRNA水平随着年龄的增加而上升 ;同一年龄组 ,甲减组较正常组nNOSmRNA表达下降。 结论提示甲状腺激素对nNOSmRNA表达有上调作用 ;Objective To investigate nitric oxide (NO) contents, nitric oxide synthase (NOS) activities and expression regulation of nNOS gene by thyroid hormone(TH) in neonatal rats of different ages. Methods Perinatal hypothyroidism was induced by infusing PTU solution to the dams (1% PTU 0.01 ml/g per day)beginning at 15th embryonic day. Biochemistry measurement and semi-quantitative polymerse chain reaction (RT-PCR) analysis were used. Results Biochemical measurement showed that both NO content and NOS activities rose with the ages, and they were lower in hypothyroidism rats than age matched controls (P<0.01). RT-PCR analysis showed that nNOS mRNA levels (corrected by GAPDH) also raised with the ages, and decreased significantly in hypothyroidism rats of different ages(P<0.01). Conclusion The expression of nNOS gene was up-regulated by TH.NO may take part in the course of impairment of brain development induced by TH deficiency.
