实时荧光PCR法与流式细胞术两种方法检测HLA-B27的比较

摘　　要：目的比较实时荧光PCR和流式细胞术两种方法检测人类白细胞抗原-B27(HLA-B27)。研究两种HLA-B27检测方法的差别和临床诊断的价值。方法分别采用实时荧光PCR法和流式细胞术两种方法检测138例病人血中HLA-B27,分析和比较两种方法的检测结果。结果两种检测方法检测结果的差异不具有统计学意义(P>0.05),两种方法的符合率为97.8%(135/138)。结论实时荧光PCR法与流式细胞术两种方法检测人类白细胞抗原-B27均准确和可靠,并且实时荧光PCR法能够判断流式细胞术所不能识别的双阳性结果。Objective To compare Real-time fluorescence PCR with flow cytometry method in detecting HLA-B27.To study the difference of the two HLA-B27 testing methods,which can offer reference for clinical diagnosis.Methods Real-time fluorescence PCR methods and flow cytometry were used to detect HLA-B27 in 138 samples.The following results were compared and analyzed.Results There was no significant difference among the two methods.The accordance rate of the two methods is 97.8%（135/138）.Conclusion The Real-time fluorescence PCR and flow cytometry are both accurate and reliable methods in detecting HLA-B27.The Real-time fluorescence PCR can determine the result accurately from the double positive（HLA-B27 ＋ and HLA-B7＋） result which flow cytometry can not differentiate.
