地圣珠水煎剂对血管性痴呆小鼠海马区NF-κB及COX-2表达的影响

摘　　要：目的:观察地圣珠水煎剂对血管性痴呆(VD)模型小鼠脑组织中核转录因子-κB(NF-κB)及环氧化酶-2(COX-2)表达的影响。方法:采用双侧颈总动脉夹闭-缺血-再灌术造成缺血再灌注脑损伤小鼠模型,随机分为假手术对照组、模型组、地圣珠组,造模后待动物清醒后当日即开始给药。地圣珠组每只小鼠按76.2g.kg-1/d灌服4g/ml的地圣珠水煎剂,模型组给予等体积生理盐水,各组给药均连续2周。ELISA法测定小鼠模型脑海马组织中NF-κB、COX-2的水平。结果:与对照组相比,模型组小鼠脑组织NF-κB、COX-2水平显著升高,而地圣珠水煎剂能明显减少血管性痴呆小鼠模型脑组织NF-κB、COX-2水平。结论:地圣珠水煎剂具有下调VaD小鼠模型脑组织中NF-κB及COX-2水平的作用。Objective： To observe the effects of Dishengzhu Decoction on the level of NF-KB and COX-2expression in hip- pocampus of mice model with vascular dementia. Methods： The synthetic vascular dementia mice model through transient occlu- sion of bilateral carotid artery were duplicated. The Chinese medicine group was administered respectively with the Dishengzhu Decoction while the model group with the normal saline only. The level of NF-KB and COX-2in hippocampus of mice model were detected by the means of ELISA method. Results： The level of NF-KB and COX-2obviously increased in model group compared with the Chinese medicine group, and while Dishengzhu Decoction could obviously decrease the level of NF-KB and COX-2. Conclusion： Dishengzhu Decoction can remarkably decrease the level of NF-KB and COX-2in hippocampus of mice model with vascular dementia.
