摘 要:目的:观察滋阴清热方对SLE阴虚内热证患者外周血单一核细胞(PBMC)蛋白表达谱的影响。方法:用滋阴清热含药血清与阴虚内热型SLE患者的PBMC共孵育,用表面增强激光解析电离-飞行时间-质谱(SELDI-TOF-MS)检测检测孵育后PBMC蛋白表达谱。结果:滋阴清热含药血清和空白对照比较,3个蛋白峰表达有统计学差异;滋阴清热含药血清和大鼠空白血清比较,1个蛋白峰表达有统计学差异。结论:滋阴清热含药血清对阴虚内热证型系统性红斑狼疮患者外周血单一核细胞蛋白表达谱的具有影响,SELDI-TOF-MS是寻找SLE患者PBMC表达差异蛋白的有效方法,结果对SLE研究具有积极意义。Objective: To observed the influence of the nourishing Yin and clearing Heat formula on the protein expression profiles of the peripheral blood mononuclear cells (PBMCs) in the patients with systemic lupus erythematosus and the TCM syndrome of Internal Heat due to Yin deficiency. Methods: 15 SLE patients with the syndrome of Internal Heat due to Yin deficiency were observed, The PBMCs of the SLE patients with Internal Heat due to Yin deficiency was incubated respectively with the mouse serum containing the nourishing Yin and clearing Heat medicine, only with mouse serum as blank serum controls group, and with RPMI1640 culture medium as controls group for 48 hours The protein profiles of the PBMCs expressed before and after co-incubation with medicated serum were detected by the surface-enhanced laser desorption ionization time-of-flight mass spectrometry (SELDI-TOF-MS). Results: The expression of three proteins in the medicated serum group was significantly lower than that of the control group (P 〈 0.05); one protein of the medicated serum group was significantly lower than that of the blank serum control group (P 〈 0.05). Conclusion: The SELDI-TOF-MS is an effective approach for seeking the nourishing Yin clearing Heat formula on the differential proteins expressed in the PBMCs of the SLE patients with Internal Heat due to Yin deficiency. This result might be beneficial to SLE research.
