摘 要:【目的】检测选择性切断腰5脊神经前根(L5-VRT)术后脊髓背角内IL-6的异常表达并评价其对运动神经损伤所致机械性痛觉过敏的影响。【方法】在不损伤感觉传入的情况下,L5-VRT制备大鼠机械性痛过敏模型,采用Western blot和免疫荧光组织化学方法检测IL-6在脊髓背角的表达水平并进行细胞定位,通过痛行为学测试观察蛛网膜下腔内注射IL-6中和抗体对L5-VRT大鼠机械性痛过敏的影响。分组:sham组、L5-VRT组,n=5;vehicle+L5-VRT、IL-6 neutralization+L5-VRT组,n=6。【结果】L5-VRT术后7 d较假手术组同侧DRG IL-6蛋白表达显著上调(P<0.001),IL-6蛋白表达的细胞类型为神经元细胞,L5-VRT术前应用IL-6中和抗体可抑制大鼠L5-VRT术后所致双侧机械性痛过敏的形成(P<0.05)。【结论】IL-6参与介导L5-VRT所致的大鼠后肢机械性痛过敏。[Objective] To test the expression of IL-6 in spinal dorsal horn and evaluate its effects on the development of neuropathic pain induced by L5 ventral root transection (LS-VRT). [ Method ] Rats mechanical allodynia model was prepared by LS- VRT. The expression and cellular localization of IL-6 in spinal dorsal horn was detected by Western blot and immunofluorescence staining. The behavioral test was used to observe the affection of the neutralizing antibody of IL-6 on the mechanical allodynia induced by L5 VRT. [ Result] The expression of IL-6 in neurons was upregulated in ipsilateral DRGs on day 7 after surgery (P 〈 0.001 ). Intrathecal injection of the neutralizing antibody of IL-6 administered 30 min before LS-VRT effectively attenuated bilateral mechanical allodynia induced by L5 VRT (P 〈 0.05). [ Conclusion ] IL-6 participates in regulation of the rat mechanical allodynia induced by motor nerve injury
