摘 要:目的 :初步探讨有机硅烷化的磁性氧化铁颗粒作为口服磁共振造影剂进行腹部磁共振成像的可能性。方法 :1测定水中和模拟胃酸条件下浸泡 2 4h的铁溶解度 ;2测定不同浓度有机硅烷化磁性氧化铁溶液的 T2 信号强度变化 ;3动物实验测定小鼠口服该溶液后 1h和 4h铁的体内分布 ;4观察家兔口服 6 0 ml有机硅烷化磁性氧化铁溶液后腹部磁共振图像和有无不良反应。结果 :1该磁性氧化铁溶液在水中和模拟胃酸条件下浸泡 2 4h的溶解度分别为小于 0 .1%和 13.4± 0 .4% ;2有机硅烷化溶液的浓度为 0 .0 1~ 10 mmol/ L时 T2 信号强度随铁浓度增加而下降 ;3小鼠口服该溶液 1h和 4h后 ,铁主要集中于胃和肠 ;4家兔口服 6 0ml 0 .1mg/ ml上述溶液后 ,腹部磁共振图像显示 ,胃中含铁部分 T2 信号强度明显减弱。家兔和小鼠喂药后分别饲养 4周和 2周 ,活动如常 ,未见不良反应。结论 :有机硅烷化氧化铁颗粒有可能用作口服磁共振造影剂 。Objective:To study thd possibility using organic silane attached magnetic iron oxide as an oral contrast agent for MR imaging.Methods:1)measuring the iron solubility of magnetic iron oxide coated with organic silane after immersing in medium HCI of pH 1.2 and distilled water for 24 h;2)measuring the MRI T 2 signal intensities(SI)of the iron oxide solution with different concentrations from 0.01 to 10 mmol Fe/L;3)measuring iron distribution in mice after 1 h and 4 h fed with magnetic iron oxide of 10 mg Fe/kg body weight;4)observing MR images of stomach region and side reactions of rabbits after taking orally 60 ml of 0.1 mg Fe/ml iron oxide solution.Results:1)iron solubilities of magnetic iron oxide after immersing in medium HCI of pH 1.2 and distilled water for 24 h were 13.4%±0.4% and less than 0.1%,respectively;2)T 2 signal intensities (SI) of the iron oxide solution were decreased with the increasing of the iron concentrations;3)iron distribution in mice after 1 h and 4 h fed with magnetic iron oxide indicated that iron mainly accumulated in stomach and intestine;4)MR images of stomach showed that the T 2 SI after taking orally 60 ml of 0.1 mg Fe/ml iron oxide solution was obviously decreased.The mice and rabbits activated as usual during 2~4 weeks after taking magnetic iron oxide and there was no side reactions.Conclusion:It is possible to use organic silane attached magnetic iron oxide as an oral contrast agent for abdomen MR imaging,but the adnimistration methed should be improved.
