复方落地生根膏对大鼠溃疡面肉芽中CTGF、MMP-2表达的影响

摘　　要：目的:研究大鼠创伤性溃疡创面肉芽组织中CTGF和MMP-2的表达,探讨复方落地生根膏促进大鼠溃疡愈合的机制。方法:大鼠创伤性溃疡模型120只,随机分为空白对照组、实验对照组和实验组,每组40只。3组分别用生理盐水纱块、生长因子纱块和复方落地生根膏外敷伤口。每组40只大鼠随机分4次,每次10只于实验的1、7、14天及皮肤愈合时取材。采用RT-PCR法检测CTGF和MMP-2含量。结果:实验组和实验对照组创面愈合时间显著缩短(P<0.05),改善瘢痕质量;CTGF表达水平在创面愈合的早期(第1、7天),3组间比较,差异有显著性意义(P<0.05),而MMP-2表达的差异主要体现在创面愈合的中晚期(第14天)。结论:复方落地生根膏可通过调节组织愈合过程中CTGF和MMP-2的表达而促进创面愈合并改善瘢痕质量,与常用的生长因子相比,具有简便廉验的特点,值得临床推广。Objective： The expression of CTGF and MMP-2 in granulation tissue in rat wound surface was observed for the exploration of the mechanism of Compound Luodi Shenggen Ointment for ulcer healing in rats. Methods： One hundred and twenty rats with traumatic ulcer were equally randomized into model control group, conventional treatment group（external application of normal saline gauze） and experimental group （external application of normal saline gauze including growth factor, and external usage of Compound Luodi Shenggen Ointment）. On day 1, 7 and 14 of the experiment and on ulcer-healing day, ten rats of each group were executed respectively for the observation of CTGF and MMP-2 expression by RT-PCR. Results： The ulcer-healing time in experimental group and conventional treatment group was significantly shortened （P 〈0.05）, and the scar was improved. The difference was significant in CTGF expression level on day 1 and 7, and in MMP-2 expression on day 14 between the two groups （P〈 0.05）. Conclusion： Compound Luodi Shenggen Ointment is effective and practical on promoting ulcer healing and improving scar quality in rats with traumatic by regulating the expression of CTGF and MMP-2.
