摘  要:目的探讨维甲酸诱导下神经管发育缺陷相关基因的表达。方法采用维甲酸喂服致神经管缺陷(NTDs)模型,通过收集提取健康与NTDs小鼠神经组织总RNA,用含有1100余个已知基因的中密度芯片比较胚胎9.5d、10.5d健康小鼠与同期NTDs小鼠神经管组织的基因表达差异,并对部分差异表达基因进行Northern杂交验证。结果有8个差异表达基因在E9.5d、EIO.5d两个时相点呈一致变化,其中NeK7、IGFBP5、ZWIO、Csf3r、PSMC6、Cdk5、Rb1在维甲酸处理后下调;Apoa-4在维甲酸处理后上调。结论多类基因参与了NTDs的发生过程;为研究正常神经胚形成的分子机制提供了有益线索。Objective To investigate the differential gene expressions in neural tube defects by retinoic acid induction. Methods This study used the retinoic acid (RA) - induced neural tube defects (NTDs) model, which extracts all the RNA of normal nerve tissues and nerve tissues in mice with NTDs. Then, using a medium density chip containing more than 1 100 known genes, a comparison of the differential gene expressions in the normal embryo and the same period NTDs neural tube organizations within 9. 5 days and 10. 5 days was carried out. Finally, this study verified some of the differentially expressed genes by Northern hybridization. Results 8 differentially expressed genes in E9. 5d, El0. 5d phases of the two - point showed consistent changes, while NeK7, IGFBPS, ZW10, Csf3r, PSMC6, CdkS, Rbl showed downward changes in dealing with RA - induced NTI)s group and Apoa - 4 in RA Addressed to deal with NTDs group showed upward changes. Conclusions Multiclass genes are involved in the process of the occurrence of NTDs. The research is helpful for the understanding of molecular mechanisms in normal neural formation.
