摘  要:目的探讨褪黑激素(MT)对慢性轻度应激性抑郁症(CMSD)大鼠海马区和脑脊液中肿瘤坏死因子α(TNF-α)和白细胞介素6(IL-6)含量的影响。方法 36只SD大鼠随机均分为MT治疗组(A组)、DM模型组(B组)和生理盐水空白对照组(C组)。A、B组采用慢性轻度应激建立CMSD模型。第7、14和21天采用Morris水迷宫实验评价大鼠智力行为变化;Western blot法检测海马区和脑脊液中TNF-α和IL-6的表达。结果 A组逃避潜伏期较B、C组缩短:第14天为(47.8±23.2)s vs. (56.4±29.4)s、(63.5±23.8)s;第21天为(22.4±13.6)s vs. (37.6±16.9)s、(30.5±13.1)s(P<0.01或P<0.05);A组第21天穿越站台次数较B组减少[(2.2±1.2)次 vs. (5.8±2.3)次](P<0.05)。A组TNF-α和IL-6蛋白表达含量较B组减少(P<0.05)。结论 MT能有效降低TNF-α和IL-6的表达,发挥抗抑郁作用。Objective To investigate the effects of melatonin(MT) on the expressions of tumor necrosis factor-a(TNF-a) and interleukin-6(IL-6) in rats with chronic mild stress depression. Methods Thirty-six SD rats were equally randomized into three groups of A(MT treated), B(model control) and C(blank control). The changes of intelligence and behavior were observed with Morris water maze test on the 7th , 14th and 21st day. The expressions of TNF-a and IL-6 in the hippocampus and cerebral spinal fluid(CSF) were detected by Western blot. Results The escape latency in group A was shorter than that in groups of B and C on the 14th day [(47.8±23. 2) s vs. (56. 44±29. 4) s and (63. 54±23. 8) s] and on the 21st day [(22.44±13.6) s vs. (37.64±16. 9) s and (30. 5±13.1) s](P〈0. 01 or P〈0. 05). The number of platform-passing in group A was less than that in group B [(2. 2± 1.2) times vs. (5.8±2.3) times] (P〈0. 05). Compared with group B, the expressions of TNF-a and IL-6 were lower in group A (P〈0. 05). Conclusion MT can play a role of antidepression in rats by decreasing the expressions of TNF-a and IL-6.
