青蒿琥酯抑制人子宫内膜癌HEC-1B细胞增殖及诱导其凋亡的机制

摘　　要：目的研究青蒿琥酯对人子宫内膜癌HEC-1B细胞的作用及其抗癌机制。方法 MTT法观察细胞毒作用,流式细胞术测定细胞凋亡率并进行细胞周期分析,RT-PCR法检测人子宫内膜癌HEC-1B细胞用药组与对照组Livin、Caspase-3mRNA表达。结果 MTT检测显示青蒿琥酯抑制HEC-1B细胞生长且生长抑制作用呈剂量-时间依赖性;流式细胞仪检测示青蒿琥酯作用使G0/G1期细胞比例上升(P<0.05),G2/M期细胞比例下降(P<0.05);对照组、5、20、80mg/L青蒿琥酯组细胞48h凋亡率(%)分别为6.64±0.19、6.92±0.28、36.42±0.77和11.77±0.58。实验组与对照组相比,5mg/L青蒿琥酯组凋亡率无统计学差异,20、80mg/L青蒿琥酯组凋亡率均有统计学差异(P<0.05);RT-PCR结果显示,青蒿琥酯组Livin mRNA表达下调,Caspase-3mRNA表达上调。结论青蒿琥酯体外抑制人子宫内膜癌细胞HEC-1B的生长;青蒿琥酯可能通过抑制Livin mRNA表达、进而增加Caspase-3mRNA表达,诱导子宫内膜癌HEC-1B细胞发生细胞周期阻滞,促进其凋亡,抑制肿瘤细胞增殖。Objective To explore the effect and mechanism of artesunate on the proliferation and apoptosis of human endometrial cancer cell line HEC-1B. Methods The proliferation change was detected by MTT assay; cell cycle change and apoptosis were measured by flow cytometry; the expressions of Livin and Caspase-3 mRNA were detected by RT-PCR before and after HEC-1B cell line was cultured with artesunate. Results Compared to the control group, artesunate significantly inhibited the proliferation of HEC-1B cell in dose- and time-dependent manners. Artesunate arrested the HEC-1B cell cycle at G0/G1 phase （P〈0.05）. Artesuate of 20 mg/L increased the apoptosis of HEC-1B. The expression of Livin mRNA was decreased while the expression of Caspase-3 mRNA was in- creased significantly in artesunate-treated group. Conclusion Artesunate can inhibit the proliferation of HEC-1B cell line in dose and time-dependent manners and arrest the cell cycle at Go/G~ phase. Artesunate can exert remarka- ble anti-tumor effect by down-regulating the expression of Livin mRNA and up-regulating the expression of Caspase-3 mRNA.
