摘  要:目的观察骨髓间充质细胞体外诱导后在异体小肠黏膜下层的生长情况,并比较其在组织工程SIS吊带和聚丙烯吊带修复重建压力性尿失禁(SUI)动物模型中的作用。方法实验采用32只正常成年雌性SD大鼠为研究对象,随机选取8只为对照组,其余为造模组。造模组大鼠行双侧坐骨神经近端横断术(PSNT)后,将其分为三组各8只:A组无吊带放置;B组(PPM组)行放置聚丙烯吊带;C组(SIS组)放置组织工程SIS吊带。造模14 d后,测定对照组和造模A、B、C三组大鼠的漏尿点压(leak point pressure,LPP),证实造模成功。术后14 d和28 d进行LPP测定,并显微镜下观察术后尿道周围组织结构变化。结果 PSNT术后14 d和28 d,PPM组和SIS组漏尿点压与对照组比较,差异无统计学意义(P>0.05),但与造模空白组相比,差异有统计学意义(P<0.05);PPM组和SIS组的最大膀胱容量与对照组和造模空白组比较,差异有统计学意义(P<0.05)。结论组织工程SIS吊带和聚丙烯吊带治疗大鼠压力性尿失禁均安全、有效,可作为治疗女性SUI的有效方法。Objective To observe the culture conditions of bone marrow stromal cells and compare its application between small intestinal submucosa and polypropylene mesh used for repairing stress urinary incontinence rat model. Methods Thirty- two female adult rats were randomly divided into four groups: control group (n = 8), the others were model group which were treated proximal sciatic nerve transection (PSNT). Then the model group was divided into blank group with no sling, PPM group with polypropylene mesh and SIS group with engineering tissue sling, and the leak point pressure (LPP) were com- pared and measured among the four groups at 14 d and 28 d after PSNT. Results LPP were significantly higher in PPM group, SIS group and control group than that in blank group, but there was no significant difference between PPM group, SIS group and control group. The maximal capacity of rats bladder was significantly higher in PPM group, SIS group than those in blank group and control group. Conclusion The effect of MDSC in SIS and polypropylene mesh used for repairing SUI rat model is good and safe, and it can treat female patients with SUI effectively.
