板蓝根总生物碱的含量测定方法研究

摘　　要：目的建立板蓝根中总生物碱的含量测定方法。方法以溶剂萃取酸性染料比色法测定板蓝根总生物碱含量,以4(3H)-喹唑啉为对照品,检测波长417.5 nm。结果 4(3H)-喹唑啉在4～24μg.mL-1范围内与吸光度呈良好的线性关系,回归方程为Y=0.023X+0.222,r=0.999 9,平均加样回收率为100.3%,RSD=2.17%(n=6)。结论所建立的总生物碱含量测定方法准确性较好,灵敏度高,重现性好,可用于板蓝根总生物碱的质量控制。Objective To establish a method for the determination of total alkaloid in radix isatidis.Methods A solvent extraction acid dye spectrophotometry method was adopted with 4（3H）-quinazoline as control.The detection wavelength was 417.5 nm.Results There was a good linear relationship in the range of 4~24 μg·mL-1 for arginine,the regression equation was Y=0.023X＋0.222,r=0.999 9,and the average recovery was 100.3%with RSD of 2.17%%（n=3）.Conlusion The established method for the determination of total alkaloid accuracy was accurate with high sensitivity and good reproducibility,and it can be used for the quality control of total alkaloid in radix isatidis.
