老年人群血浆HCY水平与MTHFRC677T基因多态性

摘　　要：目的:对血浆同型半胱氨酸(HCY)水平与N5,N10-亚甲基四氢叶酸还原酶(MTHFR)C677T基因位点的基因多态性进行分析,了解其在健康老年人群中的分布情况。方法:采用酶联免疫吸附法进行血浆HCY水平测定;采用聚合酶链反应-限制性片段长度多态性法(PCR-RFLP)对MTH-FRC677T进行基因多态性分析。结果:健康老年人群血浆HCY水平为(11.0±2.8)μmol/L,老年男、女性人群血浆HCY水平无显著性差异。老年男、女性人群中MTHFRC677T基因的CC、CT和TT基因型频率与C、T等位基因频率差异均无显著性。MTHFRC677T位点各基因型间,血浆HCY水平亦无显著性差异。结论:本试验数据可为MTHFR基因突变与HCY代谢相关疾病病因研究假设提供遗传学数据。Objective： To detect the levels of homocysteine and the genotype and allele frequency of methylene- tetrahydrofolate reductase（MTHFR） in old population in Changzhi. Methods： The level of homoeysteine was detected by enzyme-linked immunosorbnent assay （ELSIA）, and the genotype of MTHFRC677T was analysed by polymerase chain reaction-restriction fragment polymorphism（PCR-RFLP）. Results ： The plasma homocysteine level was （ 11.0± 2.8） μmol/L in old population, the difference was not significant between male and female in the plasma homocysteine level. The difference was not significant between male and female in MTHFRC677T genotype（ P〉0. 05） ,that was so in the allele frequency of C and＇T（ P〈0.05）. There were no differences in the homoeysteine levels among the different genotypes of MTHFRC677T. Conclusion：These genetic epidemiological data was useful for study of etiological hypothesis between MTHFR and diseases relatived to homocysteine metabolism.
