摘 要:目的:探讨坐骨神经横断后,microRNA-34a(miR-34a)对c-Met(肝细胞生长因子受体)的调控作用及对施万细胞增殖的影响。方法:采用Real-time PCR检测miR-34a和c-Met在坐骨神经横断后近侧端的表达变化;Edu染色法检测miR-34a对原代培养的施万细胞增殖能力的影响;双荧光素酶报告基因系统检测miR-34a对c-Met 3’UTR的直接靶向作用。结果:miR-34a与c-Met在坐骨神经横断后近侧端的表达呈负相关,能够抑制原代培养的施万细胞的增殖,并可直接作用于c-Met基因的3’UTR。结论:miR-34a可以直接靶向c-Met的3’UTR,从而通过下调靶基因c-Met的表达抑制施万细胞的增殖。Objective: To investigate the regulatory role of miR-34a on c-Met expression following sciatic nerve transection and the influence of miR-34a on the proliferation of Schwann cells.Methods: Real-time PCR,Edu staining analysis and dual luciferase reporter system were used to examine the expression changes of miR-34a and c-Met in proximal sciatic nerve segments following nerve transection,measure the effect of miR-34a on proliferation ability of primary cultured Schwann cells and identify the target role of miR-34a on c-Met 3'UTR,respectively.Results: The expression of miR-34a and c-Met appears negative correlation in proximal sciatic nerve segments following nerve transection.miR-34a inhibits the proliferation of primary cultured Schwann cell proliferation and targets the 3'UTR of c-Met gene directly.Conclusion: This study reveals that miR-34a targets 3'UTR of c-Met gene directly and inhibits the proliferation of Schwann cells by down-regulating the expression of its target gene c-Met.
