大鼠嗅黏膜和嗅球嗅鞘细胞体外分离培养比较

摘  要:目的:比较大鼠嗅黏膜(olfactory mucosa,OM)和嗅球(olfactory bulb,OB)的嗅鞘细胞(olfactory ensheathingcells,OECs)在体外分离培养中增殖和分化以及生物学特性的差异。方法:取SD大鼠OM和OB用差速贴壁法在体外分离培养OECs,扫描电镜观察两组OECs的形态,并用S-100和p75NGFR免疫荧光抗体双标记检测两组OECs在体外增殖和分化的情况。结果:扫描电镜观察到OM OECs以双极样为主;OB OECs有双极样、三极样和扁圆形3种形态,其中以双极样为主。免疫荧光双标结果显示:OM和OB组均有较多的S-100和p75NGFR双标阳性细胞,OM OECs胞体多为梭形,突起细长;OB OECs的形态多样,有双极样、三极样和扁圆形,胞体呈长梭形、圆形或三角形。两组OECs胞体面积和细胞周长相比较差异均无统计学意义。结论:OM OECs与OB OECs一样都能在体外分离培养与增殖。OM OECs与OB OECs分化成熟后的细胞无明显差异。Objective: To compare the biology property,proliferation,differentiation of olfactory ensheathing cells(OECs) derived from olfactory mucosa and bulb.Methods: In vitro by differing rates of attachment OECs were harvested from OM and OB.To observe the configuration of OECs the scanning electron microscope(SEM) were used.The proliferation and differentiation of OM and OB OECs were observed by the immunofluorescence antibody of S-100 and p75NGFR.Results: SEM showed that the morphological types of OM OECs mainly was bipolar.OB OECs have three kinds of morphological types.They are bipolar,round and tripolar.Most of them are bipolar.Immunofluorescence results showed that many S-100 and p75NGFR double labeled positive cells were observed in the two groups OECs.Most of OM OECs are bipolar cells.The shapes of OM OECs' cell bodies are fusiformis and Neurites are long.OB OECs have many kinds of types.They are bipolar,tripolar and round.The OB OECs' cell body is long fusiformis,tripolar and round.The soma area and perimeter of OM and OB OECs have no distinct difference.Conclusions: OM OECs,which like OB OECs,can be cultured and proliferated in vitro.There is no distinct difference on differentiation mature cells between OM OECs and OB OECs.
