三种非线性校准模式在糖化血红蛋白测定中的应用

摘  要:目的:探讨免疫法糖化血红蛋白合适的校准方式。方法:分别用全自动生化分析仪中常用的三种非线性校准模式对糖化血红蛋白试剂进行校准,比较不同校准模式下测定结果的差异。结果:50份样本在Logit-log4P,spline和line graph三种校准模式下测定均值分别为8.17,8.15和8.27(P=0.417);HbA1c浓度梯度测定中Logit-log4P和spline两种校准模式检测结果极其符合,比对曲线几乎重合,而line graph校准方式由于标准曲线未充分拟合在高值区和高低值区与前二者相比存在较大偏差,高值偏高,低值偏低。结论:免疫法糖化血红蛋白校准方式应选择Logit-log4P和spline为宜。Objective:To investigate the appropriate calibration mode for HbA1c immunoassay.Methods:Using three non-linear calibration modes that were common in automatic biochemical analyzer to calibrate the HbA1c reagents,and compare the difference of the test results among these three modes.Results: The mean value of fifty samples in three calibration modes,so-called Logit-log4P,Spline and Line-Graph,were 8.17,8.15 and 8.27(P=0.417).The measurement of HbA1c concentration gradient from Logit-log4P and Spline calibration modes were extremely consistent,and the comparison curves almost coincide.In the Line-Graph calibration mode,compared to the other two modes,there was a large deviation in both high-value area and low-value area,due to the standard curves were not fully fitting.Therefore,the high value was relatively too high and low value was relatively too low.Conclusion: The Logit-log4P and Spline calibration modes are more appropriate for HbA1c immunoassay.
