摘 要:将耳突后切割去头的日本三角涡虫(Dugesia japonica)分别置于16℃、18℃、20℃、22℃、24℃、26℃和28℃条件下培养,在体视显微镜下观察不同温度条件下眼点的再生情况,发现日本三角涡虫眼点再生的最适温度是22℃.通过石蜡切片对22℃培养的涡虫眼点的再生情况进行组织学研究,结果表明:涡虫去头培养第3d新生组织基部分化出视色素细胞团,随着再生进程视杯逐渐变大.In this study, temperature gradients (16℃, 18℃, 20℃, 22℃, 24℃, 26℃ and 28 C)were designed to culture the regenerating fragments of the post auricle from Dugesia japonica. The situation of the eyespots formation under the stereomicroscope were observed and found the optimal growth temperature for eyespots regeneration was 22 ℃ in dark. At this temperature, morphogenesis of eyespots during eyespots regeneration were examined by paraffin sections and the histological results showed that the regenerated tissue could be observed but there was no eyespots construction during the 1 - 2 days of regeneration. A small group pigment cells could be detected at the third day of regeneration and the small cup-shaped structure was detectable after 5 days of regeneration, and then the eyespots gradually grew up with process of the regeneration.
