益肾康颗粒对糖尿病大鼠尿转铁蛋白、尿微量白蛋白及肾比重的影响

摘　　要：目的:研究益肾康颗粒对糖尿病肾病(Diabetic Nephropathy,DN)大鼠尿转铁蛋白(Urine transferring,TRF)、尿微量白蛋白(Microalbuminura,MA)及肾重指数的影响,并观察不同浓度益肾康颗粒对糖尿病肾病的治疗作用。方法:SPF级SD雄性大鼠120只,随机选取108只大鼠,采用腹腔注射链脲佐菌素(Streptozotocin,STZ)53 mg/kg(2%0.265 mL/100g)诱导大鼠糖尿病模型。72 h后,眶静脉采血,测定血糖值,将血糖值≥16.7 mmol/L的96只大鼠随机分为模型组、西药对照组、益肾康低剂量组、益肾康中剂量组、益肾康高剂量组。未经STZ处理的12只大鼠测定血糖值后为正常对照组。西药对照组采用雷米普利灌胃。益肾康组采用不同药剂浓度灌胃。给药8周。给药6周末与8周末,用代谢笼分别收集每只大鼠24 h尿液,记录排尿量,取出2 mL×2份,-20℃冰箱保存。尿液采用生物发光检查尿微量白蛋白、采用酶联免疫法检查尿转铁蛋白。8周末取尿结束后,大鼠麻醉下摘取右肾,称重,并计算肾重指数。结果:西药对照组、益肾康低剂量组、益肾康中剂量组、益肾康高剂量组与正常组肾重指数比较具有显著差异(P<0.05),益肾康中剂量组与模型组肾重指数比较具有显著差异(P<0.05)。8周末西药对照组、益肾康低剂量组、中剂量组、高剂量组与模型组尿转铁蛋白比较具有显著差异(P<0.05)。8周末西药对照组、益肾康低剂量组、中剂量组与模型组24 h尿微量白蛋白比较具有显著性差异(P<0.05)。结论:益肾康颗粒对糖尿病肾病具有明显的治疗作用。Objective：To study the influence of Yishenkang Granules on urine transferrin（Urine transferring,TRF）,microalbuminuria（MA）and kidney weight index of diabetic nephropathy rat model and to analyze the different concentrations therapeutic effect of diabetic nephropathy.Methods：100 rats were randomly selected from 120 male SD rats of SPF level.The intraperitoneal injection of urea bacteria element（Streptozotocin,STZ） was used to induce diabetic nephropathy model.After 72 hours,the rats eye blood was taken for determining glucose levels.The rats whose blood glucose values were more than 16.7 mmol/L were randomized to model control group,western medicine control,Yishenkang Granules low,middle and high dose groups.20 rats without STZ were tested the blood glucose and even randomized to normal control group,ramipril was as the western control group.8 weeks later,4th weekend and 8th weekend,with metabolic cage,24 hours urine was collected respectively to record urine output,taking out 2 mL×2 copies,-20 ℃ refrigerator storage.Urine was tested by biological shine inspection.8th weekend,kidney weight of rats was determined.Results：The western medicine control group,Yishenkang low,middle and high dose groups and normal control group had significant difference on kidney weights（P0.05）and Yishenkang middle dose group and model control group had statistical significance（P0.05）.8th weekend,urine transferrin in The western medicine control group,Yishenkang low,middle and high dose groups and model control group were significantly different（P0.05）;MA in The western medicine control group,Yishenkang low and middle dose groups and model control group was obviously significant（P0.05）.Conclusion：Yishenkang Granules has obvious effect in the treatment of diabetic nephropathy.
