摘  要:目的研究急性冠脉综合征(ACS)患者外周血.CD4+CD28-T细胞是否表达肿瘤坏死因子凋亡诱导配体(TRAIL)以及CIM+CD28+T细胞是否通过TRAIL途径诱导人血管平滑肌细胞(VSMCs)凋亡。方法采用流式细胞术、免疫磁珠法、细胞克隆技术和Annexin-V/PI双染色法分析ACS患者外周血CIM+CD28+T细胞TRAIL表达及VSMCs凋亡。结果ACS患者外周血CD4+CD28+T细胞表达TRAIL显著高于CD4+CD28+T细胞(t=3.54,P〈0.01)。在效靶比为5:1和20:1时,CD4+CD28+T细胞引起VSMCs凋亡的程度显著高于CIM+CD28+T细胞(t=6.15,P〈0.05;t=5.21,P〈0.05)。加入TRAIL抗体阻断后CIM+CD28+T细胞引起VSMCs凋亡的程度明显减少(t=3.22,P〈0.01)。结论ACS患者外周血CD4+CD28+T细胞数量增多而且表达TRAIL,CD4+CD28+T细胞可通过TRAIL途径诱导VSMCs凋亡。Objective To study CD4+ CD28T cells -induced apoptosis of human vascular smooth muscle ceils (VSMCs) in patients with acute coronary syndrome (ACS). Methods The number of CD4+ CD28T ceils and the expression of TRAIL on CD4+ CD28-T ceils were determined by a fluorescence-activated cell sorter (FACS). CD4+ CD28"T cells were isolated using magnetic sand (MACS). These ceils were cloned with limiting dilution analysis. VSMCs were incubated with CD4 + CD28-T ceils. VSMC apoptosis was detected using 4', 6-diamidino-2-phenylindole dihydroehloride (DAPI). Results The number of CD4+ CD28-T cells in the peripheral blood of the patients with ACS was higher than those in the control group and those in the stable angina pectoris (SAP) group. The expression of TRAIL on the CD4+ CD28-T cells was significantly higher than that on the CD4+ CD28T ceUs( t = 3.54, P 〈 0.01 ). At different effector/target (E/T) ratios, the degree of VSMC apoptosis induced by the CD4+ CD28T cells was significantly higher than that induced by CD4+ CD28-T cells(t = 6. 15, P 〈 0. 05;t = 5.21, P 〈 0.05 ). Apoptosis of VSMCs was decreased when an anti-TRAIL antibody was added( t = 3.22 ,P 〈 0. 01 ). Conclusions The number of CD4+ CD28-T cells expressing TRAIL was significantly increased in the peripheral blood of patients with ACS, CD4+ CD28+T cells were able to induce ap- optosis of VSMCs through TRAIL pathway. The elevation in the number of CD4 + CD28+T cells may affect the stability of atherosclerotic plaques.
