摘 要:目的探讨在三维多孔支架环境里动态循环压力刺激对软骨细胞的作用。方法原代培养兔软骨细胞,传代扩增后接种于1.5 cm×0.5 cm×0.5 cm聚乳酸多孔(polylactic acid,PLA)支架,培养5 d后,施加0.5、1、2 N的正弦动态循环压力,频率0.017 Hz,加压8 h,以无压力组(0 N)作为对照。标本做扫描电镜及组织学切片染色,观察细胞数量、形态及排列的变化。结果在三维PLA多孔支架上,扫描电镜观察到,动态压力组细胞的增殖比无压力组更多,排列也更加有序;动态压力为2 N的时候,细胞增殖、形态比1、0.5 N及无压力组更好,贴附细胞计数与1、0.5 N及无压力组相比差异有统计学意义(P<0.05)。结论动态循环压力对于三维培养条件下的软骨细胞有促进增殖和有序排列的作用,但何种强度的压力更适合体外构建组织工程软骨尚需进一步研究。Objective To investigate the effect of dynamic compressive stress on 3-dimentional cultured chondrocytes.Methods Chondrocytes of New Zealand rabbit were obtained and proliferated on polylactic acid(PLA) porous scaffold with a size of 1.5 cm×0.5 cm×0.5 cm.The cell-scaffold composites were divided into 4 groups,and were exposed to 0,0.5,1 and 2 N dynamic cyclic compressive stress respectively for 8 hours(in a frequency of 0.017 Hz).Scanning electronic microscope was performed to examine the cell morphology on the scaffold,and histological staining was used to observe the cell prolif eration and arrangement on the scaffold after compressive stress stimulation.Results On the 3-D PLA scaffold,cells proliferated and arranged better when they were exposed to dynamic cyclic compressive stress,and 2 N group was better than 1,0.5 and 0 N groups(P0.05).The number of cells attached to scaffold in 2 N group was significantly different from that of 1,0.5 and 0 N groups(P0.05).Conclusion Dynamic cyclic compressive stress may be beneficial to chondrocytes pro liferation and arrangement on 3-D scaffold,but what extend of strength will be the best condition needs further study.
