摘 要:[目的]探讨多维分离策略从半枝莲中分离野黄芩苷的可行性。[方法]采用具有互补分离机理的大孔树脂法及有机溶剂萃取法,以及高速逆流色谱法在优化的条件下分离半枝莲中的野黄芩苷类成分,通过高效液相色谱分析检测,质谱法定性。[结果]采用D101大孔树脂粗分离时,20%乙醇水洗脱组分固形物含量较高。采用有机溶剂萃取法溶剂系统为正丁醇∶乙酸乙酯∶水(1∶1∶2,v/v/v)时,水相中主成分含量较高。高速逆流色谱法溶剂系统为正丁醇∶水(1∶1,v/v)时,能达到较好的分离效果,对主成分峰进行鉴定后确定为野黄芩苷。[结论]结合多维分离策略,能够从半枝莲中高效分离制备得到野黄芩苷。[Objective] To study the feasibility of the multiple dimensional separation method to take scutellarin from scutellaria barata D.Don.[Methods] Scutellarin was isolated by separation methods with complementary separation mechanisms under optimized condition,such as macroporous adsorption resins,liquid-liquid extraction method,and HSCCC.HPLC and MS analysis were applied to detect the separation result.[Results] Using the D101 macroporous adsorption resins,high rate of paste could be obtained with the 20% EtOH/H2O solution fraction.Liquid-liquid solvent extract method with the solvent system of n-butanol: ethyl acetate: water(1∶ 1∶ 2,v/v/v),high concentration of main compound existed in the water layer.The HSCCC separation solvent system of n-butanol: water(1∶ 1,v/v) was suitable for further separation.The main compound isolated was determined to be scutellarin.[Conclusion] High separation efficiency could be achieved with the multiple dimensional separation method.Scutellarin could be separated from scutellaria barata with the method.
