摘  要:目的:探讨蝉花菌丝对TGF-β1作用下肾小管上皮细胞α-SMA、FN、LN、ColⅢ蛋白及mRNA表达的影响。方法:以TGF-β1剌激下的肾小管上皮细胞为研究对象,采用血清药理学研究方法和免疫组化、RT-PCR检测技术,观察空白对照组、TGF-β1对照组、冬虫夏草组、蝉花高剂量组、蝉花中剂量组和蝉花低剂量组的α-SMA、FN、LN、ColⅢ蛋白及mR-NA表达,采用SPSS11.5统计软件统计分析。结果:TGF-β1剌激下的肾小管上皮细胞α-SMA、FN、LN、ColⅢ蛋白及mRNA的表达与正常组比较明显增加(P<0.01);蝉花大、中、小剂量组均能下调肾小管上皮细胞对α-SMA、LN、FN、ColⅢ蛋白及mRNA的表达,与TGF-β1对照组比较差异有统计学意义(P<0.05~0.01),并且大剂量的蝉花菌丝组优于虫草菌丝组(P<0.05);不同剂量的蝉花菌丝组存在量效依赖关系。结论:本研究表明蝉花菌丝通过下调TGF-β1刺激下的肾小管上皮细胞对α-SMA、LN、FN、ColⅢ蛋白及mRNA的异常表达而发挥抗肾间质纤维化的药理作用。Objective: To study the effect of Cordyceps Sobolifera Mycelium (CSM) on the protein and mRNA expressions ofα - SMA ,FN ,LN, Col Ⅲ in tubular epithelial ceils (TEC) when exposed to TGF - β1. Methods:The TECs were divided into normal control group, TGF- β, control group, Cordyceps sinensis (CS) -treated group, high dose CSM (HDCSM) -treated group, middle dose CSM (MDCSM) - treated group and low dose CSM (LDCSM) - treated group. The protein and mRNA expressions of a - SMA, FN,LN,ColⅢ were examined by serum pharmacological method, immunological histochemistry and RT- PCR. All statistical tests were performed with SPSS 11.5. Results:The protein and mRNA expressions of α - SMA ,FN ,LN ,Collll were significantly up - regu lated in TGF - β, control group compared with normal control group ( P 〈 O. 01 ). Treatments with LDCSM, MDCSM and HDCSM down regulated the expressions of α- SMA ,FN,LN ,ColⅢ in a dose -dependent manner, all of which were significantly lower than that in TGF- β1 control group(P 〈 0.05 ~ O. 01 ). And the expressions of α SMA, FN, LN ,Col Ⅲ in HDCSM -treated group were lower than that in CS - treated group ( P 〈 O. 05 ). Conclusion: Our study suggests that CSM may ameliorate renal tubulointerstitial fi- brosis induced by TGF - β1, which is probably attributable to normalizing the expressions of α - SMA ,FN ,LN,ColⅢin TECs.
