摘 要:目的:通过观察高糖刺激下的足细胞表达synaptopodin、desmin和Smad7的变化,探讨高糖对足细胞的损伤机制。方法:将培养的小鼠足细胞分为正常糖组、高糖组、甘露醇组。倒置显微镜下观察足细胞的形态变化,采用RT-PCR和Western印迹技术分别检测synaptopodin、desmin、Smad7mRNA和蛋白的表达。结果:高糖刺激48h后的足细胞形态发生改变,并且synaptopodin、Smad7mRNA和蛋白表达较对照组减少,desmin mRNA和蛋白表达较对照组增加(P<0.01)。结论:高糖能够诱导足细胞发生转分化,并且这一作用可能与Samd7表达减少有关。Objective :To observe the changes of expression of synaptopodin ,desmin and Smad7 by high glucose exposure in cultured podocytes and to explore the possible injured mechanisms of high glucose. Methods:Mouse podocytes were cultured and di vided into normal glucose group(NG) ,high glucose group(HG), mannitol group(MN). The cell morphological changes were traced with inverted microscope. Real - time PCR and Western blot was applied respectively to detect the mRNA and protein expression lev- els of synaptopodin, desmin and Smad7. Results:The cells converted within 48h from typical arborized cells into cobblestone appear ance induced by high glucose. The cells significantly up - regulated the mRNA and protein expression of desmin, reduced the expres sion of synaptopodin and Smad7 induced by high glucose after 48 h ( vs. NG group, P 〈 0.01 ). Conclusion: High glucose can induce epithelial -to -mesenchymal transition(EMT) in podocytes, and it maybe have a relationship with the reduced expression of Smad7.
