高碘对子1代大鼠小脑外颗粒层的影响

摘  要:目的研究高碘营养对子一代大鼠小脑外颗粒层(external granular layer,EGL)发育的影响。方法将SD大鼠40只,随机分为低碘组(0.6μg/d)、正常碘组(6μg/d)、25倍碘组(156μg/d)和50倍碘组(306μg/d),每组10只,应用MIAS-2000型图像分析系统对不同组出生后15日龄子1代大鼠小脑EGL进行形态学测量,同时对细胞核增殖抗原(proliferating cell nuclear antigen,PCNA)阳性细胞进行计数比较。结果低碘组EGL厚度[(22.3±4.6)μm]和PNCA阳性细胞率[(64.55±6.43)%]明显大于正常碘组[(16.5±3.4)μm和(45.19±8.24)%],均P<0.01;25和50倍碘组与正常碘组比较,随碘摄入量的增多,EGL厚度和PNCA阳性细胞率有增高的趋势,50倍碘组[(21.2±4.2)μm和(55.60±6.89)%]与正常碘组比较,差异均有统计学意义(P<0.05)。结论低碘和50倍碘可以延迟小脑EGL细胞的增殖,但25倍碘组未见明显变化。[ Objective] To study the influence of excessive iodine intake on the external granular layer ( EGL) of cerebellum in the first filial generation of rats. [ Methods ] A total of 40 SD rats were randomly divided into low iodine group ( SID, 0.6 μg/d), nor- mal iodine group (NI, 6 μg/d) , 25 times excessive iodine group (25HI, 156 μg/d) and 50 times excessive iodine group (50HI, 306 μg/d) , with 10 rats in each group.. The morphology of the external granular layer (EGL) and the number of cells which ex- pressed proliferating cell nuclear antigen(PCNA) of 15 days rats after birth were measured by computed MIAS-2000 Image Analysis System. [ Results] The thickness of EGL [ ( 22.3 ± 4.6 ) μm ) ] and the number PCNA ( + ) cells [ ( 64. 55± 6. 43 ) % ] in LI group was significantly higher than those in NI group[ ( 16.5± 3.4) μm], (45.19% ± 8.24 ) % ], both P 〈 0.01. All changes a- bove in HI groups showed increasing tendency with increase of iodine supply, especially in 50 HI group [ (21.2 ± 4. 2) μm, / 55.60 ± 6. 89 ) % ] were significantly higher than those of the control group ( P 〈 0. 05 ). [ Conclusion ] Iodine deficiency and severe iodine excess[50HI) can delay the generation of EGL, which does not happen to the mild iodine excess group (25HI).
