摘 要:目的通过检测关木通、广防己不同药物配伍组中马兜铃酸A的含量,探寻降低肾毒性的最佳配伍组。方法 (1)色谱柱为symmetry ODS(C18,4.6mm×250mm,5μm);流速:1mL/min;温度:室温;检测波长:250nm;流动相:乙腈-(水+冰醋酸)为58∶42(v/v)。(2)色谱柱为Diamon C18(250mm×4.6mm 5μm);柱温:30℃;流速:1.0mL/min;检测波长:316nm;流动相:乙腈-(醋酸-三乙胺)为45∶55(v/v)。结果 (1)与关木通组比较,木通茯苓组、导赤散+当归组、导赤散组、木通栀子组、木通黄连组和导赤散+黄连组的马兜铃酸A的含量分别为:0.1706、0.1402、0.1219、0.1195、0.0778、0.0442和0.0265mg/L。(2)与广防己组比较,广防己生晒参组、广防己肉桂组、广防己茯苓组、广防己生晒参肉桂茯苓组、广防己黄连组和广防己生晒参肉桂茯苓黄连组的马兜铃酸A的含量分别为:1.6830、0.4960、0.3930、0.3700、0.2930、0.1760和0.0520mg/L。结论配伍黄连后,马兜铃酸A的含量能显著降低,黄连与关木通、黄连与广防己是减毒的最适配伍。Objective To research into the optimal compatibility of decreasing nephrotoxic substance,with high-performance liquid chromatography(HPLC) method for determination of the content of aristolochic acid A in Caulis aristolochiae manshuriensis,Aristolochia fangchi after Rhizoma coptidis was matched.Methods(1)The analytical column was symmetry ODS C18 column(4.6mm×250mm,5μm),the mobile phase consisted of a mixture,acetonitrile-acetic acid 58∶ 42(v/v).The detection wavelength was 250nm.The column was at temperature room.The flow rate was 1.0mL/min.(2)Diamon C18 column(250mm×4.6mm,5μm)used as chromatographic column,acetonitrile-(acetic acid-triethylamine)45∶ 55(v/v)as mobile phase,a parameter 316nm as the detection wavelength,30 degrees centigrade selected as the column temperature and a 1.0mL/min as the velocity.Results(1)The contents of aristolochic acid A in Gmt group,GmtF group,DcsD group,Dcs group,GmtZ group,GmtH group,DcsH group were 0.1706、0.1402、0.1219、0.1195、0.0778、0.0442 and 0.0265mg/L individually.(2)The contents of aristolochic acid A in Gfj group,GfjS group,GfjR group,GfjF group,GfjSRF group,GfjH group,GfjSRFH group were 1.6830、0.4960、0.3930、0.3700、0.2930、0.1760 and 0.0520mg/L individually.Conclusion The concentration of aristolochic acid A was decreased in Caulis aristolochiae manshuriensis,Aristolochia fangchi after Rhizoma coptidis was matched.It was optimal compatibility of decreasing nephrotoxic substance that Rhizoma coptidis matched Caulis aristolochiae manshuriensis or Aristolochia fangchi.
