摘  要:目的:建立同时测定链荚豆中胡萝卜苷和β-谷甾醇含量的方法。方法:采用高效液相色谱-蒸发光散射检测法,色谱柱为Diamonsil C18柱(4.6 mm×150 mm,5μm),流动相甲醇-水(95∶5),流速1.0 mL·min-1,漂移管温度80℃,载气体积流量2.5 L·min-1。结果:胡萝卜苷和β-谷甾醇浓度分别在10~100 mg·L-1(r=0.999 3)和25~250 mg·L-1(r=0.999 1)线性关系良好;平均加样回收率分别为99.5%,98.9%,RSD分别为2.4%,2.1%。结论:该法专属、简便、准确,可用于同时测定链荚豆中胡萝卜苷和β-谷甾醇,为链荚豆的质量控制提供依据。Objective: To develop a new method for simultaneous determination of daucosterol and β-sitosterol in Alysicarpus vaginalis. Method: HPLC-ELSD method was used to determine daucosterol and β-sitosterol. The separation was performed on a Diamonsil Cla column (4.6 mm x 150 mm, 5 Ixm) with methanol- water (95: 5) as the mobile phase at the flow rate of 1.0 mL .min^-1. The temperature in drift tube was 80 ℃ and the carrier gas flow was 2.5 L .min^-1. Result: The linear ranges of daucosterol and β-sitosterol were 10-100 μg . mL^-1 (r =0. 999 3) and 25-250 mg .L^-1 (r = 0. 999 1) , respectively. The average recoveries were 99.5% , 98.9% , and RSDs were 2.4% and 2. 1% , respectively. Conclusion: This method is selective, simple, accurate, which can be applied for simultaneous determination of daucosterol and β-sitosterol in A. vaginalis.
