摘 要:目的:观察清热解毒方对2型糖尿病大鼠胰岛β细胞及骨骼肌细胞NF-κB、IκB mRNA表达的影响。方法:60只SD大鼠随机分为正常对照组、模型组、中药高剂量组、中药低剂量组、二甲双胍组,采用小剂量链脲佐菌素加高脂饮食饲养的方法建立2型糖尿病肥胖大鼠模型。给药4周后应用Real-time PCR检测大鼠胰岛β细胞及骨骼肌细胞NF-κB、IκB mRNA的含量。结果:模型组NF-κB mRNA含量较正常组显著升高,与模型组大鼠相比,中药组大鼠胰岛β细胞及骨骼肌细胞NF-κB的mRNA含量显著降低(P<0.05);模型组IκB mRNA含量较正常组显著降低,与模型组大鼠相比,中药组大鼠胰岛β细胞及骨骼肌细胞IκB mRNA表达上调(P<0.05)。结论:清热解毒方可明显抑制大鼠胰岛β细胞及骨骼肌细胞NF-κB/IκB信号通路,延缓糖尿病并发症的发生。Objective: To observe the regulating effect of Qingrejiedufang on the expression levels of NF-κB/IκB mRNA in the Pancreatic β cells and skeletal muscle cells of type 2 diabetic rats.Methods: To establish type 2 diabetic rats in obsess by injecting of low-dose STZ and fedding with high glucose and high-fat diet,after that the rats were randomly divided into 5 groups: normal group,model group,High dose of Chinese medicine group,low dose of Chinese medicine group,metformin group.After given different treatments for 4 weeks the rats were executed to test the expression levels of NF-κB/IκB mRNA.Results: Compared with model groups,the expression levels of NF-κB mRNA in the Pancreatic β cells and skeletal muscle cells of Chinese medicine group are significantly lower(P0.05),but the difference between Chinese medicine groups were not significant,the same is true amongand there is no significant difference among Chinese medicine groups and metformin group;the expression levels of IκB mRNA in the Pancreatic β cells and skeletal muscle cells of Chinese medicine group are upgrade.Compared with normal group,the expression levels of IκB mRNA are lower.Conclusion: Qingrejiedufang can significantly inhibit the NF-κB/IκB signaling pathways in pancreatic β cells and skeletal muscle cells of type 2 diabetic rats,delay the significantly inhibit the NF-κB/IκB signaling pathways in pancreatic β cells and skeletal muscle cells of type 2 diabetic rats,and delay the occurrence of diabetes complications.
