摘  要:目的:研究金耳多糖(TMP)对豚鼠哮喘模型支气管平滑肌收缩功能的影响,初步分析其作用机制。方法:采用肺阻力(RL)和肺动态顺应性(Cdyn)指标评价TMP对致敏豚鼠抗原攻击前后肺功能变化的影响;采用离体气管条法观察TMP对豚鼠气道的直接扩张和对抗氨甲酰胆碱(CCH)和组胺引起的气道平滑肌收缩作用。结果:在整体试验中,TMP 1.60g/kg预处理,能明显抑制致敏豚鼠抗原攻击后引起的RL增加和Cdyn下降;在离体试验中,5-80mg/mL TMP对豚鼠气道具有直接舒张作用,10-80mg/mL TMP对CCH引起的气道平滑肌收缩呈现非常显著的抑制作用(P<0.05),但对组胺引起的气道平滑肌收缩无对抗作用。结论:TMP抑制豚鼠哮喘模型肺功能下降可能与CCH作用有关。Objective: To observe the effects of Tremella mesenterica polysacchrides(TMP) on bronchoconstriction in asthmatic model of guinea pigs,and preliminary analysis its mechanism of action.Methods: The parameters of lung resistance(RL) and dynamic lung compliance(Cdyn) were used to evaluate the effect of TMP on pulmonary function in the sensitized guinea pigs before and after antigen challenge in vivo.We observed the effect of TMP on the resting tension of the isolated tracheal smooth muscle of guinea pigs in vitro and the contractive responses induced by carbachol and histamine.Results: The pretreatment of TMP(1.60g/kg) significantly inhibited the increase of RL and decrease of Cdyn in the sensitized guinea pigs in vivo,which were treated by antigen.5-80 and 10-80mg/mL TMP could relax the resting tension and inhibit the contraction response induced by carbachol respectively in the isolated tracheal smooth muscle of guinea pigs(P0.05),but had no effect on the contraction responses induced by histamine.Conclusion: TMP could inhibit the decrease of pulmonary function in asthmatic model of guinea pigs.It might be associated with resistance to carbachol.
