摘 要:目的:观察滋补肾精方对肾精不足证小鼠瘦素(Leptin)及瘦素受体(Ob-R)和神经肽Y(NPY)水平的影响,并探讨脾肾相互影响机制。方法:将50只小鼠随机分为正常组、模型组、金匮肾气丸组及滋补肾精方高、低剂量组。造模同时,模型组灌胃0.9%氯化钠溶液(0.02mL/g)、金匮肾气丸组灌胃金匮肾气丸混悬液(2.7g.kg-1.d-1)、滋补肾精方高、低剂量组分别以滋补肾精方(20g.kg-1.d-1、10g.kg-1.d-1)灌胃,连续21d。观察小鼠一般状况,血清Leptin、脑组织Ob-R和NPY表达水平。结果:滋补肾精方能够改善小鼠肾精不足证表现。与模型组比较,滋补肾精方高、低剂量组可明显降低Leptin及Ob-R活性,升高NPY活性(P<0.05,P<0.01)。结论:滋补肾精方可能通过调节Leptin及Ob-R与NPY水平的途径发挥补肾填精作用,脾肾相互影响是通过Leptin-Ob-R-NPY轴实现的。Objective: To observe the effect of Zibu Shenjing Fang on Leptin,Ob-R and NPY level of mice with kidney essence insufficiency,and to explore the each other influence mechanism of spleen and kidney.Methods: 50 healthy male Kunming mouse were divided into 5 groups: normal group,model group,Jinkui Shenqi Wan group,Zibu Shenjing Fang high and low doses group.With established the animal models of kidney essence insufficiency,model group was given saline(0.02mL/g),Jinkui Shenqi Wan group was given Jinkui Shenqi Wan(2.7g·kg-1·d-1),and Zibu Shenjing Fang high-dose and low-dose groups were given Zibu Shenjing Fang(20g·kg-1·d-1,10g·kg-1·d-1) respectively for 21 days.Then observed the general situation of animals,measured the level of Leptin in blood,Ob-R and NPY in Brain tissue.Results: Zibu Shenjing Fang obviously makes the symptoms of the mice with kidney essence insufficiency better,adjusts the activity of Leptin in serum and Ob-R,NPY in Brain tissue(P0.05,P0.01).Conclusion: Zibu Shenjing Fang may play an important role of Filling Kidney essence by adjusting Leptin,Ob-R and NPY,spleen and kidney influence each other is through the Leptin-Ob-R-NPY to realize.
